Acer mono Maxim,belonging to the genus Acer,is a insect-pollinated tree,mostly scattered in northeast of China.It is a roadside shade tree in many regions owing to the colorful foliage,having a high medicinal value and is as an important light industrial raw material.In order to protect and utilize this resources efficiently,in this study,we would use a combination of cpSSR and nSSR to detect the genetic diversity and population structure among 22 populations.Our aims were to: determine the extent of genetic diversity within and among populations,evaluate the degree of population differentiation and genetic structure,analyze the connection between genetic difference and climatic factors,and discuss the implication of present findings for conservation and improvement of this species in China.The outcome were described as follows:1.Based on the cpSSR data:(1)From the 23 pair primers,11 pairs of cpSSR primers were filtered.A total of 30 loci were selected.The magnitude of PPB was 97.33%;(2)Based on the magnitude of I,population in RC was highest(I=0.2672)and population in QS was lowest(I=0.0045).Based on the province of distribution,Shaanxi province was owning the richest genetic diversity,Beijing city was poorest.(3)Principal component analysis showed that: temperature and precipitation has reached higher contribution rate in nineteen climatic factors.(4)Correlation analysis showed that: this genetic diversity parameters showed a significant positive correlation with the climatic factors(P<0.05).(5)AMOVA analysis revealed that the genetic difference mainly present among populations,accounted for 82% of the total variance and only 18% within populations.(6)Based on UPGMA clustering analysis method,PCoA method and Structure clustering method,the study shows that: all studied populations in general clustered into three clade and a significant positive correlation occurred between the genetic distance and geographic distance(r=2819,P<0.01).(7)According to the characteristics of the data,we detected the kind of haplotypes was 26,the haplotype frequencies in H1 was the highest(10%),H12 and H14 was the lowest(1%);According to the geographical distribution,the haplotypediversity in Jiyuan(JY)population was the highest(22%),whereas the Labagou(LBG)population was lowest(5%).2.Based on the nSSR data:(1)From the 20 pair primers,8 pairs of nSSR primers were filtered.A total of 31 loci were selected.The magnitude of PPB was 96.44%;(2)Based on the magnitude of I,population in RC was highest(I=0.2883)and population in QS was lowest(I=0.0043).Based on the province of distribution,Hebei province was owning the richest genetic diversity,Beijing city was poorest.(3)Principal component analysis showed that: temperature and precipitation has reached higher contribution rate in nineteen climatic factors.(4)Correlation analysis showed that: this genetic diversity parameters showed a significant positive correlation with the climatic factors(P<0.05).(5)AMOVA analysis revealed that the genetic difference mainly present among populations,accounted for 78% of the total variance and only 22% within populations.(6)Based on UPGMA clustering analysis method,PCoA method and Structure clustering method,the study shows that: all studied populations in general clustered into three clade and a significant positive correlation occurred between the genetic distance and geographic distance(r=0.2525,P<0.01).(7)According to the characteristics of the data,we detected the kind of haplotypes was 22,the haplotype frequencies in H1 was the highest(11%),whereas H15 was the lowest(1%);According to the geographical distribution,the haplotype diversity in Jiyuan(JY)population was the highest(22%),the Lingkongshan(LKS)population was lowest(4%).
