Broccoli,also known as green cauliflower,is a variant of Brassica napus cabbage species,rich in protein,carbohydrates,minerals,vitamins and other nutrients.In each accounted harvest seasona,about 40-50%of the broccoli plant by-product will be abandoned in the field(including stem,leaf and residue),which will not only cause serious environmental pollution problems but also a waste of resources.In this study,broccoliresiduewill be treated as the raw material resource for animal feed production by using the fermenting process.The methologies and the processing characters are studied as the main focuses.Through theoptimized utilization of broccoli we try to provide an economical and rational use of the by-products of broccoli.].30 Strains of Bacillus were screened from 3 samples of soil.residues,and sludge from the field of broccoli in Taizhou City,Zhejiang Province,by using casein medium.And by usingthe casein medium and cellulose culture medium for a variety of experiments,a strain of bacillus,which was coded as CTL-25,was screened out asthe strong degradation of protein and cellulose ability.By using anhydrous calcium carbonate containing 2%MRS,a solid culture medium was used to screen the lactic acid bacteria strains for stronger acid producing ability in broccoli leaves samples,which was named as CTM-1.Through the identificationfrom NCKI,we found that the CTL-25 and CTM-1 strains were Bacillus subtilis and Enterococcus faecium,respectively,by 16S rDNA gene sequence alignment and homology analysis.2.Bacillus subtilis CTL-25,Enterococcus faecalis CTM-1 and Yeast CTJ-1 were inoculated at a ratio of 1:1:1,2.52x106 CFU/g in broccoli residues as 50.0%with 10.0%of cornbarn,20.0%of rice bran,and 20.0%of wheat bran of in the composition of the feed formula fermentation,the results showed that the content of acid soluble protein in the broccoliresidue was 5.19%,and the content of acid soluble protein in the feed was 5.19%the content of acid soluble protein in the broccoli residue feed was 4.98%,5.91%,4.89%and 5.29%respectively.The results showed that the compound fermentation of three kinds of bacteria had great potential in improving the content of acid soluble protein in broccoliresidue.3.The three kinds of bacteria were inoculated at 1:1:1 ratio of 5.04×106CFU/g in three different broccoli residue formulations,the content of the formula diacid was 5.56%.And then the CT combined bacteria to 2.52×106 CFU/g inoculation amount of single bacteria,three bacteria fermentation experiment,the ratio of bacteria was 1:1,the ratio of bacteria was 1:1:1,the results showed that the use of fermentative bacteria did not only increase the probiotic species of feed and the content of acid soluble protein in feed was higher than that of the other two treatments(p<0.05).The best combination of bacteria was determined by single factor experiment and orthogonal test.Ratio of CTM-1:CTJ-1:CTL-25 = is 1:14:6.4.Through single-factor experiments and orthogonal experiments,the optimal fermenting conditions was as follows:strain CTM-1,strain CTJ-1,strain CTL-25 ratio as1:14:6,inoculation amount as 7.5× 106 CFU/g,fermentation time as 3d,water content was 47.0%and the fermentation temperature was 31 ?.The content of acid soluble protein was 6.68%,the crude protein content was 23.5%,the pH value was 4.5,the reducing sugar was 3.0%.Compared with the control group without adding bacteria,the content of acid-soluble protein was increased by 150.0%,the crude protein was increased by 15.0%.the pH value was 4.5,and the reducing sugar was increased by 50.0%.
