Isolation And Identification Of Mycoplasma Ovipneumoniae

With the development of sheep industry,Mycoplasmal pneumonia of sheep(MPS)has become a worldwide disease.At present,MPS is prevalent in varying degrees in China.In recent years,MPS occurred in the sheep industry in Hohhot,which has caused greater economic losses.In this study,samples of suspicious disease cases were taken from the area of Hohhot,and isolation and identification of Mycoplasma ovipneumoniae(Movi)were carried out,in order to understand and grasp the characteristics of Movi,which has been prevalent in recent years in Hohhot.This study laid the foundation for further study on the basic research of the epidemic strains of Movi and the application of this disease.The diseased sheep lesion lung tissue was inoculated in the modified Hayflick medium,and a strain of Movi was isolated and named as Strain Movi.HS2016.The isolate was grown on solid medium with white,small colonies which was irregular round and no"fried eggs" under the microscope.The results of biochemical identification showed that the glucose test was positive,the arginine was not hydrolyzed,the urea was not decomposed,the methylene blue reduction test was positive,and the red blood cell adsorption test was positive.The 16SrRNA of Strain Movi.HS2016 was amplified using PCR and the sequence analysis was confiremed.The result showed that the homology of the Strain Movi.HS2016 was 99%all with Strain Movi FJ-FZ,Strain Movi FJ-SM,Strain Movi MoGH3-3,Strain Movi 060R03,Strain Movi XJ-3f,Strain Movi Dovre and Strain Movi Y-98 with BLAST method.The Strain Movi.HS2016 was identified by PCR using the hlyA gene of Movi.The results showed that there was a bright band at 375bp,which was the purpose fragment.The Sequence analysis showed that the homology of the hlyA gene of the Strain Movi.HS2016 was 97%with the Movi ATCC 29419 with BLAST method.
