Chrysanthemum(Chrysanthemum morifolium Ramat.),native to China,cultivated variety genus of Chrysanthemum,which has high ornamental and economic value.It is the one of ten famous traditional flowers in china and one of four most important cut flowers in the world.Our country is the center of origin and genus germplasm distribution for Chrysanthemum.Although rich in germplasm resources,cultivated of variety,color and pattern is abundant.However,genetic background and cultivated has a variety of Chrysanthemum,it is prone to variation in the production process.Color as an important ornamental traits,the study of color variation also attracted the attention of many researchers.Traditional Chrysanthemum of Chinese total of more than 3,000 varieties,more than 400 kinds of sport selection is obtained.We had the colors mutation of Chrysanthemum by selecting in the field,in variation petals as explants.Regeneration by tissue culture techniques isolated mutant of traits.Identification of the material separated by SRAP,filter specific sites which may cause color variation,lay the foundation for further exploration in the formation mechanism of color.The main results are as follows:1.Observed in the field of 12 bud mutation with color of chrysanthemum cultivars,separated by tissue culture techniques in bud mutation trait,we got 'Anastasia Green','QX-166,' 'Nannonggongxun' of chrysanthemum material bud mutation separation offspring.After observed of ' Green Anna' bud mutation separation and found that the presence of offspring color separation,may be related to different parts of the source bud mutation of petals.2.We used 40 pairs of primers were screened and isolated on the collection and preservation of 'Anastasia Green' series in mutant progeny SRAP analysis were obtained amplified bands 423,but in the 'Anastasia Green' and 8 were expanded in mutant offspring by polymorphic bands did not get specific sites cause color bud mutation,indicating color mutant of the series may involve only point mutations.Also,the primer composition may further limited relevant with we screened.3.'Anastasia White' which was bud mutant of 'Anastasia Green' offspring as the test material,establishment of an efficient genetic transformation system.In 'Anastasia White'aseptic leaf explants,we found that on MS+6-BA1.0 mg·L-1+NAA 0.5 mg·L-1 differentiation medium,which differentiation rate highest to 67.78%;to hygromycin(Hyg)as a selectable marker for genetic transformation,the results show that:8 mg·L-1 as leaf discs differentiated selection pressure is more appropriate;8 mg·L-1 concentrations suitable for the rooting screening.
