Hippophae rhamnoides L.subsp.Sinensis Rousi belongs to the species of species of Hippophae,Elaeagnaceae.It has rich bioactive substances and high medicinal value.So if we try to develop Hippophae industry,it must solve the problem of ecological governance and bring some economic value.In this study,the wild Hippophae resources in Shanxi Province were studied.For the first time,the structural characteristics of leaf cross section of Hippophae during the development of Hippophae rhamnoides L.subsp.Sinensis Rousi in Tanghe Village,Laiyuan Town,Qixian City were studied.Also,the key enzyme activities in the process of flavonoid synthesis was determined content of rutin was determined by high performance liquid chromatography.And we can explore the relationship between them.The results can provide theoretical basis for the picking time of Hippophae leaves,and can also provide guidance for the further development and utilization of Hippophae leaves.The Hippophae leaves were fixed in the middle of each month from April to October.The structural features were observed by light microscope.You can see: Hippophae rhamnoides L.subsp.Sinensis Rousi leaves only have one main vein.There are more ducts,so them have strong transport capacity.The ability of the phloem to transport organic nutrients is also strong Hippophae rhamnoides L.subsp.Sinensis Rousi leaves epidermal cells arrange closely and have very thick stratum corneum.It has more stomata,but mostly stomata is in the lower epidermis.Hippophae rhamnoides L.subsp.Sinensis Rousi leaves surface have rich epidermis hair.Epidermis hair is less and scattered in upper epidermal,and more lower epidermal.So,Hippophae rhamnoides L.subsp.Sinensis Rousi leaves have a good water retention effect.These structures are the result of long-term living in arid and semi-arid areas.Hippophae rhamnoides L.subsp.Sinensis Rousi can be used as a very good tree specie,and it is an important role to theecological environment and economic development of arid areas and semi-arid areas.The key enzyme activities in the synthesis of flavonoids from Hippophae rhamnoides L.subsp.Sinensis Rousi leaves were determined by microplate reader from April to October.The results showed that the activities of phenylalanine ammonia lyase(PAL),flavonol synthase(FLS),chalcone isomerase(CHI)and chalcone synthase(CHS)were all up and down,not obvious.Determination of flavonoid synthesis key enzyme activity,can understand the rate of synthesis of flavonoids,the regular pattern of accumulation of flavonoids and the amount of accumulation,which can provide a scientific basis for the rapid development of Hippophae resources.The content of rutin contained in the Hippophae rhamnoides L.subsp.Sinensis Rousi leaves in different months was determined by HPLC.The chromatographic column was Diamonsil C18 column(150 × 4.6mm,5?m).The mobile phase consisted of methanol-0.5% phosphoric acid(33:67),and the flow rate was 1.0 mg·g-1,the detection wavelength was 254 nm and the column temperature was 30 ?.The results showed that the content of rutin in the leaves of Hippophae rhamnoides L.subsp.Sinensis Rousi(?)was in the order of(mg·g-1): 0.31720,4922,0.4241,0.8969,0.98069,0.3869,1.4616 in April to October,The rutin content in the leaves of Hippophae rhamnoides L.subsp.Sinensis Rousi(?)was in the order of(mg·g-1)from April to October: 0.4415,0.5319,0.7300,0.9339,1.3611,0.9302 and 0.8779.As the medicinal value of rutin is higher,it can provide the basis for the further development and utilization of Hippophae leaves.
