Drosicha corpulenta Kuwana(Hemiptera,Margarodidae),is a phytophagous insect which harm a variety of trees,whose feeds juice of host plants phloem by the way of prercing-sucking.Due to ingestion of single food,it is hard for D.corpulenta to satisfied requirement.Endosymbionts could providing nutrients that limited in the host diet.These facts suggest that endosymbionts affecting the reproductive development of D.corpulenta.In this study,I investigated the type of gut microbiota and the interation of gut microbiota on the nutrition metabolism in D.corpulenta.This provides theoretical basis for new biological protection by targeting symbionts.The research content includes.(1)D.corpulenta in different instars,select two-instae larva and adult which parasitic on populus,diospyros and juglans trees.The V3+V4 hypervariable 16 S rDNA region of bacterial were amplified and sequenced by high throughput sequencing,then spliced data,divided OUT,statistical species abundance and analyzed the diversity and difference of samples.(2)Determination of nutrients in the phloem of three host plants by biochemical methods and analysis relationship of host plant,D.corpulenta and gut microbiota.Insects were fed branch that using compositic antibiotic solution to achieve the purpose of eliminate the gut microflora.By dectecting the change of nutrients in insect haemolymph,analysis the impact of gut microbes on insect nutritional metabolism.Result as follows:1.Different host plant and different ages of D.corpulenta’s intestinal microorganism diversity and difference.The resules showed that Bacteroides and Proteobacteria are the dominant bacteris in two-instae larva on populus,diospyros and juglans.Bacteroides,Blattabacteriaceae,Candidatus Uzinura respectively accounted for 79%,81%,94%.Proteobacteria,Enterobacteriaceae respectively accounted for 20%,6%,6%.The resules also showed that Bacteroides,Proteobacteria and Actinobacteria are the dominant bacteris in adult insect on populus,diospyros and juglans.Candidatus Uzinura respectively accounted for 72%,59%,92%.Wolbachia respectively accounted for 0.01%,12%,0.01%.Candidatus Lariskella respectively accounted for 25%,0.001%,0.001%.Enterobacteriaceae respectively accounted for 12%,0.2%,1.1%.Arthrobacter respectively accounted for 0.7%,1%,2.4%.Alpha diversity analysis showed that adult higher than two-instae larva on diversity index.The general level has annotated nearly 70 kinds of bacteria through high-throughput sequencing.Two-instae larva intestinal dominant bacteria in the genus is Candidatus Uzinura and Enterobacteriaceae.Adult intestinal had added Wolbachia,Candidatus Lariskella,Arthrobacter nearly 27 species.Skermanella are unique to populu adult.Kluyvera and Paracoccus are unique to diospyros two-instae larva.Tsukamurella,Enterobacter,Brevundimonas,Streptococcus has percent of 0.15% were relatively more in juglans adults.2.Nutrients in the phloem of three host plants and effects of changes in intestinal microflora on host nutritional metabolismExperimental measured populus,diospyros and juglans trees phloem of total sugar,reducing sugar,protein,and tannin content,the highest content of suger.D.corpulenta can get suger and energy required from plant.Diospyros and juglans trees has hightest content of tannin that plant defense insects.However we discovered the bacteria digestive tannins through the analysis,to break the plant defense to D.corpulenta get normal parasitic life.We use antibiotics to clear bacteria in the gut,and determine the content of glucose,triglyceride and albumin in insect hemolymph,and the result indicate that the increase in glucose and triglyceride and the decrease in albumin are one of the important reasons for bacteria loss of intestines.This experiment studied the gut microbiota of different hosts and different ages,and the results showed that the dominant bacteria,the structure of the population and the prediction of bacterial function,and confirmation the gut microbiota affect nutrient metabolism of D.corpulenta.This study provide theoretical basis for determine the advantage bacterium and the host protein utilization,which provides new thinking for the development of biological control technology.
