8-Oxoguanine DNA glycosylase 1(OGG1)is a key enzyme which recognizes 8-oxoG and initiates the base-excision repair mechanism to excise 8-oxoG.It is an evolutionarily conserved gene whose function is repairing the oxidative DNA damage caused by the reactive oxygen free radical(ROS)and preventing the occurrence of mutagenesis of 8-oxoG.Mutation of OGG1 in human leads to tumors;AtOGG1 is related to seed maturation and germination,influences the longevity of seed.There have been no reports of OsOGG1.There were overexpression?RNAi?promoter vector transgenic lines of OsOGG1 before this research.We constructed vectors for genome editing and subcellular localization of OsOGG1,screened the already existing transgenic lines and obtained the homozygous lines,conducted a series of resistance experiments on overexpression,RNAi homozygous lines and to explore whether OsOGG1 has the function of resistance to biological and abiotic stress.All these results will provide gene resource and theoretical supports for improving the rice resistance ability.Detailed results are as follows:1.Obtained overexpression ? RNAi ? GUS promoter GFP promoter homozygous transgenic lines.The OsOGG1 T0 genome editing transgenic strains and got 5-8 yet homozygous mutants with significantly declined expression level of OsOGG1.2.The OsOGG1 subcellular localization and stable vector pYBA-1132-OsOGG1 was constructed and the genetic transformation of rice(in the process of tissue culture)was carried out.3.The preliminary results of rice blast and bacterial blight resistance showed that OsOGG1 overexpression and RNAi transgenic lines did not showed a certain more resistant or susceptible phenotypes than wild type.4.The expression level of oOGOG1 has a certain response to JA treatment.The expression of OsOGG1 showed a decreasing trend when JA was treated in Japan for 1 h,and decreased slightly at 6 h.After treatment for 12 h,the expression of OsOGG1 decreased sharply.5.The seeds of Nipponbare?OsOGG1 overexpression?RNAi and genome editing yet homozygous transgenic lines were artificially aged and then were conducted a germination test.The results showed that the germination potential and germination rate of all the treated seeds decreased with the increase of aging time.After the same aging time,the germination potential and germination rate of OsOGG1 overexpressing seeds were higher than that of Japanese Significantly higher than OsOGG1 silencing and OsOGG1 genome editing of transgenic seeds.
