Dolang sheep as the research object of Qimubulake breeding cooperatives in Kashi.Motivated by mining major candidate genes and verification,the study is to perform whole genome resequencing to explore genes across Duolang sheep genomic,and get candidate genes by means of statistical analysis with biological information.Then we disscused two candidate genes' SNPs associated with reproductive traits of Duolang sheep.This study would design a unique molecular marker for the characteristic variety in Xinjiang Province,and laid the foundation for the selection of high density molecular marker assisted selection.The results as follows:(1)Using Illumina Hiseq2000 platform to sequence each multi lamb group and single lamb group with mixed pool.There were 13.8M SNPs be found in multi group and 13.6M SNPs be found in single group.Based on those SNPs,using Fst,gene heterozygosity and genome-wide association(GWAS)to detect makeed SNP location.Combined with ENSEMBLE database and David online to get candidate genes.At last,we got 6 candidate genes associated with reproductive traits,they are NCOA1?INHBA?ING5?BMPR-IB?ARNT?KLHL1.(2)Designing primers to find possible SNPs location of products and make a test on 168 Duolang sheep blood.Acrossing analysis each SNPs that associated with litter size,we got those results:Two SNPs was found at 7 exon of NCOA 1 gene,there are three genotypes in 95 bp location:CC,CT,TT and the C/T mutation caused the change of proline.Analysis of the relationship between polymorphism of two SNPs and litter size that shows there is no difference between them.We also found three SNPs at 18 intron and 20 intron,acrossing analysis of the association between polymorphism of 3 SNPs and litter size,the study showed the SNPs effects of NCOA1 gene on litter size are not significant.We only found a T/G mutation at 1 exon of INHBA,analysis of INHBA genotypes and litter size relationship with least-square method that shows the differences are not significant,too.
