| Drynariae rhizoma is the traditional Chinese medicine with a relatively long history,initially recorded in the A Supplement to Materia Medica.Due to its reliable efficacy,Drynariae rhizoma has been praised by ancient physicians,and is still used today.Drynariae rhizoma has the effect of replenishing the kidney,strengthening the bones,promoting blood circulation,hemostasis and relieving pain,also can be used to treat deficiency syndrome of kidney,marked by back pain,tinnitus,impairment of hearing and looseness of teeth,traumatic injury,bone fracture,external use for alopecia areata,vitiligo.Modern pharmacological researches show that Drynariae rhizoma has the obvious pharmacological activity against bone and joint disease,cochlear toxicity and skeletal development and so on,which proves that it is a promising traditional Chinese medicinal materials of high economic benefits.With the development and utilization of a large number of Chinese herbal medicine,the phenomenon of market chaos occurs frequently due to insufficient supervision.However,for a long time,Drynariae rhizoma,as a commonly used traditional Chinese medicine,because of the similar character and efficacy,and the regional prescribing habits and etc,are often replaced with its adulterants.So the Drynariae rhizoma varieties are chaotic.At present,there are few reports on the authentication of Drynariae rhizoma and its adulterants,and most of the studies focus on the macroscopic and microscopic authentication.There is no systematic study on the authentication of Drynariae rhizoma and its adulterants,which will restrict the quality and development of the medicine.Therefore,this subject will be on the Drynariae rhizoma and its adulterants for pharmacological studies,and the establishment of DNA barcoding.In this article,first of all,the pharmacognosy study of Drynariae rhizoma and its confused species was completed.1.Origin Authentication: Determining the origin of Drynariae rhizoma and its adulterants through the investigation;2.Macroscopic Authentication: Summarizing the characteristics by observing its appearance;3.Microscopic Authentication: To observe the microscopic characteristics of the scales leaves and transection,and summarizing the morphological characteristics of the scales and the arrangement vascular bundles of transaction;4.Physicochemical Authentication: Improved the thin-layer chromatography(TLC)of Naringin in Drynariae rhizoma,and established a new TLC method of Protocatechuic acid in Pseudodrynariae rhizoma,we can identify Drynariae rhizoma and its adulterants combined with the two TLC results.The content of Naringin in Drynariae rhizoma was determined by HPLC.The results showed that the concentration of Naringin showed a good linear relationship with the peak area when the concentration of Naringin was in the range of 5.01~86.14 μg/m L.The method of characteristic chromatogram of Drynariae rhizoma was established by High Performance Liquid Chromatography(HPLC).Comparing with the HPLC chromatograms of its adulterants,the chromatographic’s behavior of Drynariae rhizoma was significantly different,which can distinguish between Drynariae rhizoma and its adulterants.DNA barcoding approach was used to identify Drynariae rhizoma and its adulterants from gene level.The established DNA barcoding of Drynariae rhizoma and its adulterants were based on the sequence of psbA-trnH chloroplast genome,and investigated the genetic relationship among Drynariae rhizoma and its adulterants.Based on the four major authentication methods,this paper completed the characteristic chromatogram of Drynariae rhizoma,established the DNA barcoding of Drynariae rhizoma and its adulterants,and comprehensively improved the systematic study of the authentication of Drynariae rhizoma and its adulterants and enriched the quality standard and solved the problem of authentication of Drynariae rhizoma in the actual market circulation.Meanwhile,this article provides a new idea for the pharmacognostical research of Drynariae rhizoma,lays the foundation for improving the quality control standards of Drynariae rhizoma. |
