Objectives 1.To establish high-yield culture technique of Rhizoma Amorphophalli in Zhejiang;Formulate production cultivation techniques of the best germination rate and production.2.To optimize the extraction procedure of konjac glucomannan3.To select the parts with best effect through comparing the different extracts of Rhizoma Amorphophalli4.To select the optimum conditions in anticancer through analysis of dose-effect and time-effect relationship.Methods 1.To set different plant density,lateral corm size,planting date and interplanting mode,keep the record of the plant height,the emergency rate and the yield and so on and select the optimum cultivation technique.2.3,5-2 nitro salicylic acid(DNS)colorimetric method is used to determine the content of konjac glcomannan,Examine the impact of each factor by univariate method,and orthogonal design is used to optimize extraction processes of konjac glucomannan.3.To extract from Rhizoma Amorphophalli decoction with petroleum ether,ethyl acetate and n-butyl alcohol,divide the different extracts into 4 parts,test the anticancer activity through Oxford cup method and the optimum extracts and concentration.4.To explore the effect of different extracts of Rhizoma Amorphophalli on the proliferation of the cell of stomach cancer and compare the anticancer activity.Results 1.April 20th is the optimum plant date,200g is the optimum weight,2000 plant/mu is the optimum density.2.With the increase of the extraction rate of the solid-liquid ratio also increases,the maximum extraction rate when in the solid-liquid ratio of 60:1,followed by the extraction rate decreases slightly;with increasing leaching time,KGM extraction rate also increased,after 3h time with increasing extraction rate increases very slowly;in the range of 30??40?,as the temperature increases,a corresponding increase in the rate of extraction,the extraction rate sharply after 40?.3.Antibacterial activity intensity for each component of petroleum ether component>ethyl components>butanol components>water component.Antibacterial activity of the strongest components of petroleum ether.4.The petroleum ether and ethyl acetate extracts have effect on the cell of MKN45 which have dose-effect relationship and the n-butyl alcohol extract and water decoction doesn't work in this experiment which have nothing to do with time.Conclusion 1.The yield of Rhizoma Amorphophalli is effected by the lateral corm size and the plant density mostly above all.2.Orthogonal experimental study of these three factors,the effect of temperature on the extraction rate of the largest,followed by solid-liquid extraction time is over again;the best system for the extraction of KGM:solid-liquid ratio of 60:1,extraction time 3h,extraction temperature 40?.3.Site of the strong possibility of antibacterial activity in petroleum ether component among its minimum inhibitory concentration of 3mg/mL.4.The petroleum ether and ethyl acetate extracts are the effective parts which have anticancer activity.
