Anti-inflammatory Chinese Medicine "Gong Yanjing" Compound Optimization Research

Object:Extracting the total compound alkaloid,simplified compound alkaloid and alkaloid of Leonurus japonicas from self-prepared traditional Chinese medicine "GongYan Jing" and researching the invitro bacteriostatic action of this alkaloids on the main pathogenic bacteria(staphylococcus aureus,streptococcus and Escherichia coli)of cow endometritis,the anti-inflammatory experiments adopted mouse ear swelling model,toe swelling model,celiac capillary permeability experiment,and granuloma inflammation model to evaluate the anti-inflammatory effect of each treatment.,Provide the basis for optimizing the self-prepared traditional Chinese medicine "GongYanJing".Methods:1.Crude extract self-prepared traditional Chinese medicine "GongYanJing"(leonurus japonicas,ligustici,angelica,peach kernel,liquorice,Faeces Trogopterori,dried ginger,cattail pollen)added iodine potassium iodide reagent and bismuth potassium iodide reagent,silicotungstic acid reagent,phosphomolybdic acid reagent,the detection of each medicine whether it contained alkaloids,the preliminary screening of alkaloid containing each drug was refined,the alkaloid containing single leonurus japonicas,ligustici and liquorice compound composition simplified.2.Through ethanol refluxing extraction and ultrasonic assisted extraction were extracted from traditional Chinese medicine "GongYanJing" alkaloids,simplified compound alkaloid and alkaloid of Leonurus japonicas.3.Researching the invitro bacteriostatic action of this alkaloids on the main pathogenic bacteria(staphylococcus aureus,streptococcus and escherichia coli)of cow endometritis by using agar diffusion plate method and tube twice times dilution method.4.Through the establishment of mouse ear swelling model,toe swelling model,celiac capillary permeability experiment,and granuloma inflammation model to evaluate the anti-inflammatory effect of each treatment.Results:1.Join the Chinese medicine alkaloid reagent "GongYanJing" in 8 kinds of single herb extracts,found leonurus japonicas,ligustici and liquorice sinensis contain alkaloids,the alkaloid containing single leonurus japonicas,ligustici and liquorice compound composition simplified.2.Compared with ethanol refluxing extraction and ultrasonic assisted extraction method showed that the ethanol reflux extraction of Chinese herbal compound "Gong YanJing" compound,and simplify the monarch drug in Herba Leonuri alkaloid extraction rate is higher,the extraction rate of ethanol reflux extraction is 0.92%?0.93%?0.71%,Ultrasonic assisted extraction is 0.87%?0.86%?0.62%.3.Researching the invitro bacteriostatic action of this alkaloids on the main pathogenic bacteria(staphylococcus aureus,streptococcus and escherichia coli)of cow endometritis by using agar diffusion plate method,the average inhibitory circle diameter is 17.4±0.8mm?16.9±1.2mm?7.8±0.8mm,The above three groups of alkaloids on Staphylococcus aureus MIC,MBC were 32 g/L?125 g/L?500g/L,62.5 g/L?125 g/L?500g/L;Escherichia coli MIC and MBC were 62.5 g/L?125 g/L?250g/L,125 g/L?250 g/L?500g/L;The MIC and MBC of Streptococcus lactis were 125 g/L?125 g/L?500g/L,250 g/L?250g/L.4.The mouse anti inflammation test found that the traditional Chinese medicine compound "GongYanJing",and the simplified compound motherwort alkaloids on mouse ear swelling,foot swelling and celiac capillary permeability and granuloma formation has good inhibitory effect,the compound "Gong YanJing" and simplified compound anti-inflammatory effect is better than that of motherwort(P<0.05),no significant difference between the two groups(P>0.05),simplified the anti-inflammatory effects of compound the compound is slightly better than the total compound "GongYanJing".Conclusion:Self-prepared traditional Chinese medicine "GongYanJing" antibacterial effect in vitro and in vivo anti-inflammatory activity and the simplified compound had little difference,provides a theoretical basis for the clinical application of simplified compound.
