Avena Fatua L.is an important gene bank for improving wheat genetic resources, which has the advantages of tolerant to stress, resistant to diseases, and high nutrition. The progeny from wheat × hexaploid wild oat were investigated in order to exploit fully the excellent genes of wild oat by genomic in situ hybridization?GISH?and fluorescence in situ hybridization?FISH?in the present study. The main results were as follows. The results of C-banding and FISH analyses with oligo-?GAA?7 probe in T.timopheevii showed that FISH pattern by Oligo-?GAA?7 probe was consistent with the C-banding pattern. But oligo-?GAA?7 signals were about 20% less than those of the C-bandings.Oligo-?GAA?7 probe could accurately identified 11 pairs of chromosomes in T.timopheevii.Oligo-pAs1-1 and Oligo-pSc119.2-1 could distinguish all the chromosomes of A and G genomes.Translocation line TG F4-4-2 with whole arm from Avena Fatua L.Var.glabrata Pat was selected successfully in the progeny from wide cross wheat × Avena Fatua L.by GISH.The results showed that a pair of arms of Avena Fatua L.Var.glabrata Pat were located in the 4A chromosome. This translocation was also found in next generation plant F5-4-2,indicating that this translocated arms could be transmitted steadily to their offspring. This wheat-Avena Fatua L.translocation line would have a significant value for research and application.
