In this paper,we analysed the nutritional utilization tendency of 32 strains of L.edodes,thus screen out the strains which could more suitable to cultivate with crop straw.Use maize and sorghum straw partly replace the sawdust as the culture substrate,and according the results of agronomic characters of L.edodes to screen out the formula.And 6 mutant strains were obtained by 60Co-? ray,these strains suitable for the cultivate in crop straw.So,we can fully utilizing straw resources,saving woody resources and cost.1.Preliminary screening of L.edodes strains with straw utilization abilitiesThe 32 strains of L.edodes were cultured in CMC-Na medium and xylan medium respectively,No.7 preliminarily screen out the utilization abilities of various strains to cellulose,hemicellulose and straw.Then fruiting experiments to determine whether they can steadily fruiting.According to the screening results,the No.1 and795 strains were have better utilization abilities for cellulose and hemicellulose,the No.7 strain was better than other strains utilization ability for cellulose,the No.1?Shen9?Zhenzhu?No.6?2015111202 and Cr04 strains have better utilization abilitiy for hemicellulose.After the fruting results,we found many strains could not suitable for coltavite in crop straw,and have higher contamination rate.Finally,we choose the wuxiang1 as the experiment strain to screen the formula of straw.2.Preliminarily breeding of L.edodes strains suitable for cultivate in straw by60Co mutagenesisThe research purpose of this paper is to screened out the L.edodes cultivated strains which could utilization straw more effectively.In this study,the Lentinus edodes strain Qing 20 was irradiated by 60Co-? ray,and the mutagen dosage was respectively 0.4 k Gy,0.6 k Gy,0.8 k Gy,dose rate: 10 Gy/min.The mutant strains were identified by antagonistic reaction and SSR molecular marker,and preliminary screening of utilization ratio of cellulose and hemicellulose.The results showed that all strains were alive after treat with 0.4 k Gy,the death rate was 46.7% after treat with0.6 k Gy,and the death rate was 100% after treat with 0.8 k Gy.Optimal mutagenesis condition :0.6 k Gy,10 Gy/min.Six mutant strains were screened out by antagonistic reaction and SSR,and the utilization ratio of cellulose and hemicellulose were betterthan CK.3.The screening of straw cultivation formulaTo make the subsequent formula more targeted,analysis was made of the components of the sawdust?oak flour?,sorghum and corn straws and the difference in the content of lignin,cellulose and hemicellulose.It was found that sorghum straws had the highest hemicellulose;also it showed higher cellulose content than oak flour and corn straws;however,it was poor in lignin.The corn straws were relatively poor in lignin,cellulose and hemicellulose;however,they had higher lignin content than sorghum straws.The strain “ Wuxiang 1”,with desirable fruiting performance,was used as tested strain.Culture medium was made with corn and sorghum straws which partially or wholly replace the sawdust.And different amount of lignin was added to the medium to prepare 4 groups of culture medium.Besides,different proportion of tannin was added to the pure sawdust medium to prepare 3 groups of culture medium.The pure sawdust medium served as control group?CK?.There were 12 groups of culture formula in total.According to the fruiting results,the corn straw was used as the major cultivation materials,No.2 medium achieved relatively desirable effect.In other words,the additive proportion of 36% sawdust,36% corn straw and 8% lignin achieved better effect in the weight of single fruiting body,total output and biological conversion ratio than CK.Where sorghum straws were used as the major cultivation materials,No.6 medium,performed the best in fruiting effect.That was,the additive proportion of 44% sawdust,30% corn straws and 6% lignin achieved relatively good effect in total output and biological conversion ratio.Also there was good performance in the number of fruiting bodies,the weight of single fruiting body and the appearance of L.edodes.When tannin was added,No.9 medium performed the best.That is to say,the addition of 1% tannin to the original sawdust medium achieved desirable effect in the output of fruiting body and biological conversion ratio.Unexceptionally it performed better than CK in the number of fruiting bodies,the weight of single fruiting body and total output.
