According to the productiong status of Lentinan which was high cost, low yield and high price, the study on selecting of lentinula edodes strains with high polysaccharide production was conducted by ultraviolet ray mutation and monospore hybridizationg. The results as follows:1.Choosed nine Lentinula edodes strains which were cultivated widely in different area. Cr-04 and L-26 were selected as the parent strains by determined the polysaccharide production.Owned 204 cross combination which had clamp connection of edible fungi through single spore isolation,ultraviolet ray mutation and single spor crossbreeding.2.The cross combination which mycelium slow growth or weak had been reject,and others would be antagonize with parent strains.50 strains had no antagonistic streak and 56 strains only had weak streak;98 strains had significant streak were new cross combination.3.98 new crossing progenies strains were obtained by microscope checking and antagonistic experiments.37 new crossing progenies strains which binuclear mycelial are thick and white were obtained by comporation and analysis of binuclear mycelial status.4.After the comporation and analysis of polysaccharide production and stability,X19, X29,X42,X77,X185,X246,X525 were obtained which polysaccharide were more higher than parent strains.Mensurate the polysaccharide production of 7 cross combinations on 9th,10th,11th,12th and 13th,the result 11th is the fastigium.The seven strains were obtained by means of vertical slab polyacrylamid gel electrophoresis and analysis of esterase isozyme of crossing parents and new cross combination, which had mutually complementary enzyme bands and new enzyme bands.5.The effects of Carbon Sources and Nitrogen Sources for polysaccharide Production of Lentinula edodes were studied.The results showed that Starch is the best for polysaccharide Production among the tested Carbon Sources and the concentration is of 6%. The best Nitrogen Source is Yeast extract and the concentration is of 0.4%.
