| This study is aimed to use metabolomics methodology to comparative analysis the differences between antibiotics and probiotics on piglets.A total of 15 male and 15 female weaning piglets,requires similar weight in good health 28 days old were selected and divided into 5 groups: group A,group B,group C,group D,group E.Each group had six replicates with one piglet per replicate.The piglets in group A were fed the basal diet.The pigs in other groups fed the diets as followed: group B: basal diet + 1.8‰yeast culture,C: basal diet + 1.6 ‰ Lactobacillus plantarum culture,D: basal Diet + compounded culture(0.8 ‰ Lactobacillus plantarum culture + yeast culture 1.8 ‰),E: basal diet + 1 ‰antibiotics(chlortetracycline).Both of the Lactobacillus plantarum culture and Yeast Culture were dry weight.During the experimental period,feed and water were provided ad libitum.After the experimental period,all of the pigs were killed and the serum,digesta of ileum and cecum were analyzed by the GC-MS machine.The data was analyzed by the orthogonal partial least-squares(OPLS-DA)of SIMAC13.0 and t test,screening out the different substances of the metabolites.Trial 1.The effects of different probiotics on serum metabolites in piglets.The results showed that there were most kinds serum metabolites.The peak value of fructose in group A was significantly higher than that in group C and E(P <0.01).The peak value of glucose,ornithin and leucine in group A were significantly higher than that in group E(P <0.05).The peak value of prolin and valine in group A were significantly higher than those in group D and E(P <0.05).The peak value of carboxylic acid in group A was significantly higher than that in group D(P <0.01)).Trial 2.The effects of different probiotics on ileum metabolites in piglets.The results showed that eight kinds of different variables were detected in the ileum of group B(basal diet + yeast culture),and the peak of mannose was significantly higher than that in group D and E(P <0.05);9,12-octadecadienoic acid,Lactic acid and anti-9-octadecenoic acid in group D were significantly lower than group B(P <0.05).The peak value of lactic acid in group B was very significantly higher than that in group C(P <0.01).The peak value of cholesterol in group B was significantly higher than that in group A(P <0.05).The peak value of glycerol in group B was very significantly higher that in group C,D and E(P <0.01).The eight different variables in group B mainly involved in glycolysis and tricarboxylic acid recycling pathway.Trial 3.The effects of different probiotics on cecum metabolites in piglets.The results showed that nine kinds of different variable were detected in the cecum of group E(basal diet + antibiotics).The peak value of fructose was significantly higher than that in group B(P <0.05).The peak value of 9,12-octadecadienoic acid was significantly higher than group B(P <0.05).Compared with group E,the peak value of mannose in group A and B were significantly lower(P <0.05).The arabinose,xylose and galactose in group B was significantly lower than those in group E(P <0.05).The peak of inositol in group E was significantly higher than that in group B(P <0.01)and group A(P <0.05).The antibiotics had the biggest influence on cecal metabolites than other groups.In summary,the experimental group to add yeast culture is the most appropriate on the ileal,serum regulation,add antibiotics have the greatest impact on the cecal metabolic activities.Therefore,in the piglet feeding stage,it is suggested that the antibiotic effect is significant when the piglets are stressed,and the yeast culture can improve the quality of the animal in the daily feeding process. |
