Slash pine is China's important planting tree species currently,which is grows fast,strong resistance,fiber length,good materials and high fat,etc.However,The quality and reproductive capacity of slash pine seedlings is an important prerequisite for the success of large-scale afforestation.In this study,Slash Pine 10 family seed imported from the United States as the research material,Seeding and seedling culture in the Jingdezhen Maple Hill Forest Farm and Xiajiang seed field,and determination of biomass and growth of different families year seedlings to analyze variability and genetic characteristics of each character,and explore the tissue culture propagation techniques,to enhance the superior families of slash pine seedling reproductive capacity and carry out early selection to provide theoretical basis and technical support.Rresults are as follows:(1)it is significantly different that diameter,height,root length,root width,lateral root number greater than 5cm,each member of biomass accumulation,distribution ratio and stems root ratio of slash pine 10 families between Jingdezheng and Xiajiang,and biomass accumulation of Xiajiang greater than the Jingdezheng's.Seeding traits significant difference among families of high-yielding slash pine in Jingdezheng,and Seeding traits significant difference among families of high-yielding slash pine in Xiajiang except seedling height.Seedling traits degree of variation is between 2.53% and 16.64% among families of slash pine,which the most abundant of degree of variation was the number of lateral roots longer than 5cm in Jingdezheng.Family heritability of traits of seedlings was between 0.164 and 0.896,In addition to low family heritability of seeding height,the other traits of family heritability were higher than 0.5,at above average levels.Correlation analysis results of each trait showed that : it is significantly positive correlation between the Root length and the number of lateral root longer than 5cm,and the correlation coefficient was 0.723.it were not significant negative correlation between the root pieces and other traits as well as height and diameter,and It showed no significant positive correlation between each other traits in Jingdezheng area.In Xiajiang area,Between the root pieces and other traits were significant positive correlation;it were not significant positive correlation between the diameter and main root length as well as the diameter and the number of lateral root longer than 5cm,the correlation coefficient was 0.702 and 0.660;Among other traits was not significantly positive correlated.According to cluster analysis and integrated height and diameter to choose fast-growing slash pine family,The results showed that No.1,3 and 7 family is optimal.Slash pine seedlings of annual biomass accumulation from small to large are: root <stem <leaf;Dry weight of seedlings on the ground was between 16.96 g and 24.92 g,The range of average total dry weight of seedlings was 23.64g-33.8g;The range of variability of each elements of biomass,the distribution ratio and stems root ratio was 6.00%-30.10%;The variability of each elements of biomass accumulation and stems root ratio were higher than distribution ratio of each elements;Family heritability(in the rang of 0.56-0.85)of each elements of biomass,distribution ratio and stems root ratio of annual seedlings were above the average level,which was Subject to strong genetic control.(2)Using zygotic embryos of slash pine as the explants material to induce adventitious buds,the results showed that: zygotic embryos Are cultured in medium of GD+6-BA4.5 mg/L+NAA0.05mg/L for three weeks,then were transferred to hormone-free medium of GD,which is good for slash pine adventitious buds.Hormone-free medium which was added 0.5g/L activated carbon or 0.05mg/LNAA is in favor of adventitious buds elongation.Pinus elliottii aseptic seedling without roots were applied as explants to induce axillary buds and plant regeneration,The results show: The best medium type of slash pine adventitious buds is GD,and The best hormone concentration ratio was 6-BA4.5mg/L+NAA0.05mg/L;Slash pine adventitious buds soaked in DCR liquid medium which was added 40 mg /L6-BA for 18 hours,then were cultured in DCR hormone-free medium,which is good for improving the added rate of adventitious buds;The best rooting medium of slash pine adventitious buds was WPM+IBA2.0mg/L+NAA 0.05mg/L;The difference of adventitious buds,value-added and rooting ability of high-yielding slash pine among families was significant;The highest rate of adventitious buds was No.9 family,and the induction rate is 92.27%;The highest rooting rate and added coefficients was No.5 family,Respectively,64.11% and 5.55.Using zygotic embryos of slash pine as the explants material to induce callus,the results shows: the best medium for callus induction of slash pine was DCR + 6-BA0.5mg /L+NAA0.5mg /L+2,4-D2mg/L.Light blocking culture and adding 3g/L inositol in culture is in favor of postponing callus browning time and reducing the degree of callus browning,However,adjusting the sucrose concentration had no effect on callus browning.
