Correlation Analysis Of Polymorphisms Of Lactation Traits Candidate Gene With Part Of Lactation Traits In Jiangyue Donkeys

In this study,Jiangyue Donkeys were used for the main experiment object.PRLR,DGAT2 and CSN3 gene were chosen as main candidate genes.Genetic polymorphisms were detected by PCR-SSCP and DNA sequencing technique.Selected the gene of Lactation performance in Jiangyue Donkeys,found the association with PRLR,DGAT2 and CSN3 genes on the Lactation performance.Provide basis for preliminary studies and technical support for further exploration of the genetic mechanism of Lactation performance Jiangyue Donkeys breeds from the molecular level.The results were as follows:1.PRLR,DGAT2,CSN3 gene polymorphism detected in Jiangyue donkeysIn this study,the genetic polymorphism in flanking area of PRLR,intron 3,exon 5,6 of DGAT2 and exon 1 of CSN3 was detected by using PCR-SSCP mathod.Genetic polymorphism was found in PRLR-P1 and PRLR-P2,there were both 2 genotypes in this gene:AA and AB genotype,and no mutations in DGAT2 and CSN3.Gene frequency of Allele A was 0.942,0.921 respectively,allele B was 0.058,0.079 respectively,Dominant genotype was A,and both.in the Hardy-Weinberg equilibrium.Sequencing result showed that the SNP of PRLR-P1 was g.29764526 A>T.It was a nonsense mutation that created a premature stop codon that results in a truncated protein;The SNP of PRLR-P2 was g.29764584C>G,it was a Synonymous mutation,did not cause amino acid changes.2.Association analysis of polymorphism of PRLR gene with lactation traits in Jiangyue donkeysThe result of association analysis of mutation with lactation traits showed: AB genotypes were higher than AA genotypes of Average daily milk yield in PRLR-P1(P<0.05).AB genotypes were higher than AA genotypes of Average daily milk yield(P<0.05),and AA genotypes were higher than AB of protein percentage in PRLR-P2(P<0.05).The flanking SNPs of PRLR was associated to the lactation traits,and might be the important marker on lactation performance in Donkeys.
