Aphis gossypii Glover he destructive insect pets in the agriculture,horticulture and forestry,are capable of reproducing asexually and sexuall upon environmental change,spanning all regions in china.Cotton aphid can be divided in two phenotype type: Winged aphis and wingless aphid.Aphids can spread dozens of virus,and so produce honeydew plants infected with sooty.Cotton aphid has a very special life cycle.We called the non-genetic diversity.That is,the cotton aphid can selectively reproduce sexually and asexually according to changes in external conditions.Cotton aphid eggs overwinter in the winter,the following spring,sunshine growth,the temperature rises,the eggs hatch becomes dry mother,fundatrix several generations of parthenogenesis produce fundatrix.Fundatrix can produce winged viviparous female aphid migration to cotton,cucumbers and other summer host,constant parthenogenetic viviparous,breed winged and wingless viviparous female aphid.To the late autumn season,the temperature decreases,exposure time becomes short,winged masterbatch produced cotton aphid,migration to the overwintering host,wingless female aphids produce without wings male sexual female aphids produce winged male host in the summer,migration to the overwintering host,male and female mate,lay eggs overwinter in the winter host.Now Under laboratory conditions We can successfully induce GP.The condition of PF is 16l:8d 30?,the condition of GP is 8d:16l 18?.However the molecular basis of aphid reproductive mode switch remain an enigma.The insulin signaling pathway is a conservative signaling pathway,and plays a key role in regulating insect life,body size,embryo development,rhythm and wing shape.The insulin receptor is an upstream component on the IIS pathway and is a transmembrane receptor that triggers cascade of binding insulin.First insulin receptor cotton aphid-specific dsRAN was designed.Then,RNAi of insulin receptor gene in cotton aphid was conducted by injection the aphid with dsRNA of insulin receptor mixed with water,protine and entranster-in vivo,and the effect of RNAi were detected by real-time quantitative PCR.1)Compare the expression pattern of insulin receptor in G0,G1,G2 and G3,the significant expression is in G2.But the expression pattern of insulin receptor in G2 shows that the lowest expression is in G2 2nd and 3rd.2)The injection of dsIn R1 mixed with water was more significant than that mixed with protine.The ovary of G2 didn't show the diffierent phenotype.3)The result of qRT-PCR shows that he injection of dsIn R1 mixed with entranster in G1 was suppressed effectively,especially in G1 11 d.60% reduction over 24 h injection.Also after injection the ovary of G1 was inhibited.In this paper we explored new ways of RNAi in cotton.Find out the most feasible way to make the RNAi efficiency stable.The maturation of the interference technique also fills the gaps in the mode of interference of cotton aphids.
