The utilization of astragali resource are often limited to the astragali root,while the astragalus stem and leaf has always been discarded greatly,causing serious waste of traditional Chinese medicine resources.Astragalus stem and leaf also contains rich active ingredients,such as flavonoids,saponins and astragalus polysaccharides,etc.FGM strain was isolated from chicken intestine.In this study,astragalus root,stem and leaf was fermented with FGM,bacillus subtilis,and bacillus subtilis respectively.The aim is to investigate the main active ingredient including flavonoids,saponins,and polysaccharides volume changes of astragalus root,stem and leaf fermented by probiotics strain.(1)Astragalus root,stem and leaf were treated with FGM strain fermentation,respectively.The effect of fermentation on the astragalus root,stem and leaf was analyzed by comparing the main active ingredient including flavonoids,saponins and polysaccharides.The results showed that the polysaccharides and total saponins were increased after fermentation,the crude polysaccharide rates of astragali root,annual stem,two years stem,annual leaf,two years leaves were 177.46%,227.27%,207.11%,170.61% and 207.11%;Total saponins increaser rates were 68.50%,55.91%,55.71%,40.93%,55.71%.The flavonoid was also increased in astragalus root,annual stem and two years stem,the increased rates were 55.67%,33.68% and 30.04%,while this composition was reduced in astragalus leaf.This indicated that polysaccharides,saponins,flavonoids in astragalus root and stem could improve by FGM fermentation,which helps the further utilization of different parts of astragalus resources.(2)With two step fermentation,bacillus subtilis was used for preliminary fermentation on astragalus,and the extraction of first products was treated with FGM strain.The results showed that the polysaccharide content in astragalus root,annual stem,two years stems,annual leaves,leaves for two years increased after fermentation,increase rate were 93.41%,156.05%,132.51%,123.81%,121.63% respectively.This treatment could improve the crude polysaccharide content in the fermentation production,however,the increase rate of the main composition content was not obvious compared with the single use of FGM strain.This result was probably related to the incomplete extraction of astragalus polysaccharide in the first step fermentation,which needs further research.(3)The parameters were optimized under the similar to the indusdry conditions.The results showed that the FGM was cultivated for the fourth generation,and the last cultivation time was sustained 6-8 h,the fermentation temperature was 39 ?,inoculation amount was 5%,p H was 5.4,the incubation time was 48 h.Under these conditions,the increase rate of polysaccharide in fermentation production could reach to 240%.This result suggested that the extraction of polysaccharides in astragalus root could be significantly improved by FGM fermentation.(4)The dynamic trend in the polysaccharides content and the CFU number of FGM were measured from 0 h to 72 during fermentation on astragalus.Results showed that FGM increased gradually in 0-36 h,and reduced gradually after 36 h.Polysaccharide concentrations increased in 0-24 h and decreased after 24 h,and increase gradually within 24-48 h,then reduced gradually after 48 h.This result suggested that the polysaccharide was expended during the bectria growth,which leaded to reduction of the in polysaccharide reduction,after then,the cellulose was secreted from FGM,this reaction helped to imcrease the crude polysaccharide in fermentation tank.
