In recent years,with the continue-developing of the traditional Chinese medicine in our country,the company engaged in the production of patent Chinese medicine and extraction of Chinese Medicine.At present,the industrial treatment of Chinese medicine slag mainly by incineration and buried.It not only caused the environment pollution but also the waste of resource,that because the slag extracted were still contain much activity components.Therefore,somebody wanted to transfer the slag of Chinese medicine into animal feed additive,it may had the function including prevention and cure for livestock and poultry diseases,promote animal growth.Large quantities of cellulose contained in the slag were difficult to be digested by no-ruminant.Microbial fermentation technology not only could reduce the content of cellulose,improve the utilization of cellulose but also could solve the problem of the treatment of the slag.Thus,In order to offer the technical support for Astragalus slag applied in the animal husbandry,the best fermentation conditions of Astragalus slag was explored in this study.Methods: Enzyme activity of 11 strains including fungal and bacterial which could produce cellulose was measured based on the previous study.Screening the strain by measuring enzyme activity every 24 h during the 5 days of continuous culture.Then combine the screened strain,and measure the enzyme activity of all the combination by the same method.Determined the single or combine strain which had high enzyme activity,and study its effect on the amount of the Astragalus IV.The single factor investigation experiment including fermentation time,temperature,carbon source,carbon source concentration,inoculum amount and the nitrogen source concentration.The orthogonal experiment including 4 factor including nitrogen source concentration,carbon source concentration,inoculum size and fermentation time,every factor were set 3 levels.At last,the optimal solid fermentation conditions of Astragalus slag was determined by the orthogonal experiment including 3 factor including the amount of the medium,the fermentation temperature and the time of fermentation,every factor were set 3 levels.Results:(1)Two Aspergillus niger and 3 Bacillus strains which had highly active in producing cellulase activity were attained by the above strain screening.The combination of Aspergillus niger H-2 and Bacillus Y-1 had the highest cellulase activity among all the combinations.(2)The result of the amount of the Astragalus IV indicated that the amount of the precipitation of the Astragalus was positively correlated with the activity of the fiber enzyme activity,and the best strain promote the precipitation of the Astragalus saponins was Aspergillus niger H-2.(3)The results of the optimized test for cellulase production showed that the optimum conditions for producing cellulase by Aspergillus niger H-2 were that the fermentation temperature was 28℃,initial pH was 5.0,proportion of wheat bran to straw was 1:3,carbon source concentrations was 3%,nitrogen source concentrations was 0.2%,inoculation amount was 5%,fermentation time was 72 h.The solid fermentation conditions of the Astragalus slag were that the water content was 60%,fermentation time was 5 days,fermentation temperature was 28℃.Conclusions: Aspergillus niger H-2 can significantly promote the precipitation of the Astragalus saponins in Astragalus slag,and degradate the cellulose content,and it was the best strain for the fermentation of Astragalus residues..
