In this experiment,the study on the purification and rejuvenation of the pleurotus ostreatus was carried out by using issue separation,cutting-edge separation,protoplast regeneration technology with "99" strains as material.A total of 59 isolates were isolated and 19 dominant strains were obtained after preliminary screening.11 strains were obtained by identifying and analyzing the physiological,biochemical,authenticity and agronomic characters.The main results are as follows:(1)Four strains were obtained using tissue separation technology.The results showed by determining the mycelium growth speed and enzyme activity that the biological yield,laccase activity,the growth rate of mycelium of 4 tested stains were superior to that of the control strain.There were no significant differences between the4 strains in esterase isozyme and ISSR electrophoresis.The comprehensive agronomic characters were screened to obtain three rejuvenate strains by named as “PoZ1”,“PoZ2”,“PoZ3”,respectively.(2)Separation of 3 isolates by cutting-edge mycelium separation technique,through the determination of the mycelial length and laccase activity of 3 strains and the control strains,the results showed that 3 strains were superior to the starting control strains in laccase activity,fermentation broth biomass yield and biological yield,etc.3strains of the compound strain,The average growth rate of the mycelia of cultivated species was faster than that of the starting control strains.The average growth rate of 2strains was faster than that of the control strains in the original mycelia.There were no significant differences between the 3 strains in esterase isozyme and ISSR electrophoresis,and the comprehensive agronomic characters were screened to obtain two heterosis strains named " PoJ1 " and " PoJ2 ".(3)The protoplast regeneration technique was used to isolate 50 strains,and 50 strains were selected to obtain 12 heterosis strains,and the mycelium length,laccase activity and other characters of 12 complex strains were further studied,which showed that The average daily growth rate of the mycelium and the original hypha of 10 strains was significantly better than that of the control strains,the 6 strains were better than the control strains in the daily average growth rate of the mycelium,and 12 of the strains were better than the control strains in laccase activity,mycelium biomass yield and thefirst mushroom yield.There was no significant difference between the 12 strains in esterase isozyme and ISSR electrophoresis and the control strains,the comprehensive agronomic traits were screened to obtain 6 heterosis strains,named "Po11","Po23","Po26","Po30","Po45","Po46",and "Po45".The strains were superior to other strains in the long speed of hyphae,the biomass production of fermented liquid and the yield of the first mushroom.(4)The selection of strains obtained from three kinds of rejuvenation techniques were compared.The results showed that using protoplast regeneration and rejuvenation technique to purify Pleurotus Ostreatus was the best,and using protoplast regeneration and rejuvenation technique to obtain the strain in the long speed of hyphae,laccase activity,the yield of first-rate mushroom,agronomic characters and so on,it is better than the separation of tissue and the advanced separation and rejuvenation technique.
