China is a big agricultural country,agriculture and animal husbandry output value accounts for much of China's gross national product ratio, since China's accession to the WTO, all walks of life have been rapid developments, and with the development of animal husbandry has also exposed our country is the nationa short of feed resources.Asearly as the 19th century new energy and raw material feed had been developed vigorously overseas, and seaking substitute for protein feed and developing new protein feed be the the top priority. Thereby the crude protein content of straw and the palatability of straw will be improved. Moreover, manure is using to make matrix, so the pollution caused by animal manure may be improved to some extent.First of all, this pilot test screened out 17 heterologous strains from the collected 28 stains. And then using fuzzy comprehensive analytical method compared them by the dry weight, day long fast, crude protein content, growing to obtain the object of hybrid parents. By monocyte strains hybridization method derived hybrid strain of A0713. Then planting A0713 in optimal matrix, and making comprehensive analysis in the nutrition indicators of hybrid strains and 17 heterologous strains to determine the good species and fiting matrix for stains protein feed production.The results of experiment:1) First to screen the collected 28 hyphas and initially screening out 17 heterologous strains for test to ensure the heterologous, and then using fuzzy comprehensive analytical method preliminarily study the superior and inferior of 17 hyphaes, more reasonable and more objective analyzed the 28 hyphas from the dry weight, day long fast, crude protein content and growing.2) By time of enzyme treatment, enzyme treatment temperature and enzyme concentration choosed two heterologous strains. Hybrid this two kinds heterologous hyphaes, after microscopic examination, we found age of 5 days, their protoplasts at 28℃, using 1.5% lywallzyme,in 3 hours to process,we can got release of 3.6×107/mL,and then protest and the antagonism test between different strains.Though microscope test;antagonistic test with parents; fruiting of strain A0713, can basically determine the success of integration.3) Through the different carbon and nitrogen sources optimizating the medium for strains A0713, analysising the amount of gypsum added. on three levels full test using the type and percentage of added nitrogen and gypsum content showed that mushroom strains can grow in the right amount of chicken manure and cow dung added. Due to the chicken manure to add nitrogen 6% the hypha is growing significantly higher than the comparison, but when the humidity is 65%, chicken manure above 12%, hypha growing very slowly, do not thrive, not be covered in training materials. Add cow dung in the matrix at the humidity of 65%, its adding 15% showed an advantage from the protein content, growth rate, laccase activity, hyphae thick can be judged as a reasonable formula.4) In comprehensive assessment of the performance the A0713 hybrid strain is the best among 18 different strains, but the Hei Pingwang highest in the crude protein content, and Ao Hei has the top cellulase activity.
