The GUS expression vectors containing BrCYP79B2-1/2/3 and BrCYP79B3 promoter ofChinese cabbage were used for the transformation of model plant Arabidopsis thaliana.The expression profiles of BrCYP79B2-1/2/3 and BrCYP79B3 at different growth stages or after treatments of different hormones were analyzed in transgenic plants.It will be useful for the study of functional differentiation of BrCYP79B2/B3 genes in indole glucosinolate and IAA synthesis pathways.The main results are as follows:(1)The GUS expression vector of pBI-B2-2 containing BrCYP79B2-2 promoter of Chinese cabbage was constructed.pBI-B2-2 and pBI-B3 were transformed into Arabidopsis wild-type Columbia by floral dipping method.By using GUS histochemical staining method,their expression patterns were analyzed in different tissues of homozygous transgenic Arabidopsis thaliana plants at different stages.It was suggested that the expression of BrCYP79B2-1/2/3 and BrCYP79B3 was mainly concentrated in apical meristem,hypocotyls,cotyledons,true leaf veins,roots and root tips,where are the main sites of auxin synthesis and transport.So,BrCYP79B2-1/2/3 and BrCYP79B3 may not only affect the synthesis of indole glucosine,but also were related to the synthesis of auxin.In addition,the expression pattern of BrCYP79B2/B3 changed with the growth and development of plants.(2)BrCYP79B2/B3 were transformed into myb34myb122 and myb51myb122 mutants of Arabidopsis thaliana.By using GUS histochemical staining method,it is showed that the expression of BrCYP79B2-1,BrCYP79B2-2,BrCYP79B2-3 and BrCYP79B3 might be related to transcription factor MYB51,besides,the expression of BrCYP79B2-1 may also related to MYB34.(3)After treating with gibberellic acid,salicylic acid,methyl jasmonate,abscisic acid,ethylene and auxin,the expression pattern of BrCYP79B2-1/ 2/ 3 and BrCYP79B3 were analyzed by GUS histochemical staining.With 400 ?M ET treatment,CYP79B2/B3 cannot express,the expression of BrCYP79B2-1/B2-3 may be related to abscisic acid,auxin and salicylic acid.The expression of BrCYP79B2-2/B2-3 may be related to naphthyl acetic acid and the expression of BrCYP79B3 may be regulated by other factors.
