| Hexaploid wheat(Triticum aestivum L.)is one of staple food crop in the word,d urum wheat(Triticum durum)is the material for macaroni in weatern countries,Both domestic and international studies on the evolution and domestication of wheat are hig hly valued.Einkorn wheant,emmer wheat and common wheat in Triticum have wild s pecies(or primitive cultivars)and cultivars,which are extremely rare materials for the study of plant domestication and polyploification.The domestication of wheat is great significance to the development of human history.The study of key gene in the dom estication of wheat can help people to9 know more about the domestication of crops.Triticum urartu,Triticum monococcum,Triticum dicoccoides,Triticum durum,Triticum spelta and Triticum aestivum six speceies were sequenced in this study,to explore the domestication of wheat through SNP sites and differences expression levels,the result s are as follows:(1)Take the Triticum urartu as an example to evaluate the quality of Trinity asse mbly.Statistics the results of blastn found that: the vast majority of reference CDS se quences can be found similar sequences in the results of Trinity assembly,but contain a lot of dissimilarity with reference sequences in the sequences of Trinity assembly.(2)By analyzing the SNP sites of cultivated species relative to wild species,it w as found that the SNP sites was different between chromosomes and sub-genome.In T riticum durum,the number of SNP in the 2B chromosome is the most,the number of SNP sites of the 4A chromosome is the least,and the number of SNP sites in the B sub-genomes is more than A sub-genome.In Triticum aestivum,the number of SNP sites of the 1A is the most,the number of SNP sites of the 6D is the least,and the number of SNP sites decreased successively in the B sub-genome,the A sub-genome and the D sub-genome.Then,statistics SNP sites distribution on each chromosome wit h 2 M for unit 1 M for step,found SNP sites distribution on chromosome exon area also has great difference.Through annotate the gene has SNP sites of Triticum mono coccum,Triticum durum,Triticum aestivum,found annotated matabolic pathway has a large intersection of 69.95%,64.38%,100.00% respectively.(3)By examining expression difference gene between Triticum wild species and c ultispecies,expression difference gene of Triticum monococcum is 1.96% of all einkorn wheat genes,expression difference gene of Triticum durum is 10.4% of all emmer wheat,expression genes difference gene of Chinese spring is 10.4% of all common wh eat genes,in the process of gene annotation detect high expression gene and low expr ession gene of Triticum monococcum,Triticum durum,Chinese spring is annotated the same function,this phenomenon in Chinese spring is more clearly. |
