Euzophera pyriellaYang,a genus of lepidoptera,is found in many areas of Xinjiang.In recent years,with the development of Xinjiang's characteristic forestry industry?Pyrus spp,Ficuscarica,etc.?,the expansion of cultivated area,cultivated varieties,and some unreasonable agricultural measures,the damage of Euzophera pyriella has been aggravated year by year,which has become a important constraint to the development of characteristic forestry in Xinjiang.The research aimed at the sample of 13 Xinjiang geographical populations of E.pyriella,based on two mitochondrial genes of COI and ND5,using specific primers for PCR amplification,sequencing and analysis of these.Compared with the genetic diversity,phylogeny,genetic structure,and population history of different geographic populations of E.pyriella,the first study of 13 different geographical populations of P.grandiflora was conducted.The main conclusions were as follows:?1?In this study.658 bp COI gene 552,39 COI gene in the haploid type,the content of nucleotide A+T?69.33%?is far higher than that of G+C content?30.67%?,with 28 polymorphism loci,accounts for about 3.19%the total number of nucleotides.?2?557 ND5 gene fragments were obtained from 651bp,and 17 single times of ND5 gene.Similarly,there is an obvious a-t bias,with 18 polymorphic sites,accounting for 2.76%of nucleotide total.?3?The haplotype diversity,nucleotide diversity and average number of nucleotide differences in the COI gene of E.pyriella were 0.838,0.00107,and 3.481,respectively,and the haplotype diversity and nucleus of the ND5 gene.The average values of the glycosylate diversity and the average number of nucleotide differences were 0.815,0.00090,and 2.451,respectively.?4?The molecular variation analysis?AMOVA?found that the inter-individual variation of COI gene and ND5 gene in the population accounted for 94.15%and 95.43%of the total variation,respectively,which was far greater than the variation among groups and between populations.The inter-individual variability in the population of the cantharidin population dominated.According to the size of the genetic differentiation factor FST,it was found that the genetic differentiation among most geographical populations was not significant,and there were frequent gene exchanges between some populations.?5?The Mantle correlation test results of the two genes in the 13 E.pyriella geographical populations involved in this study all showed that the genetic distance between the geographical populations and the correlation coefficient between the geographical distances between populations were very low.The correlation was not significant,so it was concluded that the genetic differentiation between E.pyriella was not caused by geographical segregation,and the long-distance migration of E.pyriella was probably due to the transport of fruit and seedlings.
