Toona sinensis,which belongs to Meliaceae,has a long history of cultivation in China,and spreads around China.The tend leaves of T.sinensis is a popular vegetable,and it's also used for gardening and greening.It's leaves and seeds contain many bioactive substances,so T.sinensis also had important medicinal values which deserves deep excavation and development and utilization.In this study,93 germplasms of 10 populations collected from Shandong,Anhui,Hebei,Sichuan,Hubei,Yunnan provinces were used as materials.Its above-ground 15 agronomic traits were evaluated in order to understand characteristics of these germplasmes.ISSR,SRAP and EST-SSR markers were also used to analysis genetic diversity and genetic relations of these T.sinensis germplasms,thus helped to the resource utilization and breeding of T.sinensis.The main results were as follows:1.Evaluation of 15 above-ground phenotyping traits showed that these 93 germplasms showed plenty of character variations,and possecesed different kinds of variations.There is a significant correlation between the different characters and the different characteristics between the different populations.2.Based on the above-ground phenotyping traits,these 93 germplasms can be divided into three groups.The first groups includs 65 germplasm resources,mainly the materails collected from Hengshui,Nanjing,Chengdu,Dezhou and so on.This group characterized by thicker compound leaf petiole,longer compound leaf and larger opening angle.The second group includs 8 germplasms,mainly are materials from Taian.This group was characterized by short compound leaf petiole,short compound leaf length,short leaf length and small spacing of leaflets.The third groups includs 20 germplasms,mainly materials fromTaihe.This group was characterized by darker bud scale,partial brown,yellow light red compound leaf petiole,red leaflets petiole,longer compound leaf and little number of leaflets.3.Three kinds of molecular markers applied in this study showed high amplification efficiency in differen populations of T.sinensis,which could be used as an effective marker for the study of T.sinensis resources.The average allele number,expectation heterozygosity,Shannon information index and Nei gene diversity were all higher in 10 T.sinensis populations,indicating that the genetic diversity of Chinese T.sinensis resources was abundant.Among the 10 populations,the genetic diversity of populations from Taihe,Taian and Wuhan were higher,the lowest genetic diversity from Quanzhou population.The genetic identity among the 10 populations was high and the genetic distance was small.4.Clustering results based on the genetic identity of populations were conducted by using ISSR,SRAP and EST-SSR markers.The results showed that SRAP and EST-SSR clustering results are basically identical,except the results of Nanjing populations.ISSR clustering results were different from the results of SRAP and EST-SSR markers.Based on the clusting results of three kinds of molecular markers,these 10 populations could be divided into 3 groups.The first group consist of Hengshui,Taian and Taihe populations,the second group consist of Chengdu,Wuhan,Kunming and Ruyang populations,the third group consist of Dezhou and Quanzhou populations.There was no significant correlation between the genetic distance and the actual geographical distance of the 10 populations.5.According to the genetic differentiation coefficient of T.sinensis materials,the vast majority of genetic variation occurs mainly in the interior of populations.There are certain degree of gene exchange between different populations of T.sinensis,but not frequent.
