| Rice is one of the three major food crops in the world.Almost half of the people regards rice as food.However,Drought is the important limiting factor in rice production.Improving rice resistance to drought is one of the most important goals in rice genetic improvement.The cysteine-rich peptide(CRP)is one of the most important short peptides in plants.it play an important role in plant growth,disease resistance,plant-bacteria symbiosis and other processes.it has founded that the OsDT11 gene is a CRP that encodes 88 amino acids.OsDT11-Overexpression can significantly increased drought resistance in rice.after drought treatment,This is the first time in the world that CRP-like plant peptides have been involved in drought resistance.They have high research value in drought resistance and production practices.Based on the previous results,we performed transcriptome sequencing of wild-type and OsDT11-overexpressed plants after drought treatment to study the molecular mechanism of OsDT11 regulating ABA synthesis.and mapping the signal pathways of OsDT11 regulating rice drought resistance.using site-directed mutagenesis and tobacco transient transformation to analyzed the functional domains and key regulatory sites of OsDT11.Obtain the following research results:(1)Analysis of RNA-seq data demonstrated that OsDT11 short peptides is related to ABA to regular drought resistance in rice It is further proved that OsDT11 short peptides are involved in ABA signaling pathways,thereby increasing rice resistance to drought.(2)Three downstream target genes that may interact with OsDT11 were screened in the transcriptome sequencing data by fluorescence quantitative PCR.(3)Established and validated the system of reporter gene driven by gene LOC-Os01g73170 promoter can work with OsDT11 in tobacco transient expression system.(4)Demonstrated that the third cysteine site is the main functional site of OsDT11 by mutating the 8 cysteine sites of OsDT11 driving the reporter gene expression.In summary,this experiment further explained the relationship between OsDT11-mediateddrought-resistance and ABA signaling pathways through transcriptomic analysis,identified the key cysteine sites of OsDT11 function,and established an in vitro interaction system.The study provides a theoretical basis for further analyzing the new shortcomings of CRP short peptides in regulating drought resistance and creates new varieties of drought-resistant rice through genetic engineering. |
