The receptor for activated C kinase 1?RACK1?is a protein that exists in prokaryotes and eukaryotes widely,which has the ability of the binding activity with protein kinase C?PKC?.The protein can participate the process of the signal transduction,cell growth,adhesion and survival.Therefore,it has the role of regulation,shuttle and integration.The mytilus coruscus were mainly distributed in the coastal of China,Japan and the Korean Peninsula.With the development of the social industrialization,the pollution of the warter resources became worse and worse,and the production of mytilus coruscus was decreasing.In this paper,the receptor for activated C kinase 1?RACK1?gene was cloned.Meanwhile,the expressions of RACK1 in the mytilus coruscus under Lipopolysaccharide challenge,copper iron stress,B[a]P exposure were analyzed respectively.Due to fill the vacancy of RACK1protein in the mytilus coruscus,which provide theoretical basis for the coping ability of mytilus coruscus under stimulation with external factors to increase the yield.The results as follow:A full length 1143 bp gene encoding M.coruscus RACK1?the new gene named‘Mc-RACK1',Accession No.MG197987?was firstly cloned from cDNA library.97 bp and 92 bp nucleotide fragments were cloned as 5?untranslated region?UTR?and 3?untranslated region?UTR?,respectively.An open reading frame?ORF?of 954 bp encoding a putative protein of 317 amino acid residues with a calculated molecular mass of 35.0 kDa and pI of 7.60 was characterized.Homology comparisons showed that the deduced Mc-RACK1 had the highest sequence identity of 95%with Mya arenaria RACK1,followed by 93%identities with Pinctada martensi RACK1homolog.The evolutionary status of RACK1 in mytilus coruscus is in agreement with its biological classification,which reveals its strong conservatism.The most common conserved structure of RACK1 gene is seven tryptophan aspartate?WD?repeats.The RACK1 gene of mytilus coruscus is also predicted seven WD repeats,which revealing that RACK1 gene family has a conservative function and roes.In the In the phylogenetic tree,it is found that the mytilus coruscus were first clustered with the branches of mollusks,which conforms to the traditional taxonomy and phylogeny.The expression of RACK1mRNA in digestive gland,haemocytes and gill in mytilus coruscus under Lipopolysaccharide challenge and Cu2+stress;hepatopancreas in mytilus coruscus under benzopyrene exposure were both detected by using Q-PCR.The results showed that the expression of Mc-RACK1mRNA in different tissues were significantly increased under the stress of Lipopolysaccharide,Cu2+and benzopyrene.The high expressions of RACK1 gene of mytilus coruscus in this kind of immune related tissues reveals its participation in the defense and immune regulation in response to external stimuli.
