| Isatis indigotica Fortune is a herbaceous plant in the cruciferous plant Isatis,which root is Isatidis Radix,with the effect of clearing heat and detoxifying,cooling blood and regulating pharynx.This study is based on field trials and laboratory tests.The Isatis indigotica Fortune was studied and quality standards for seeds and medicinal materials were formulated.This study provides scientific basis for standardized production of Isatidis Radix.The results are as follows:1.From the aspect of sampling,clarity,germination rate,weight,moisture content,viability and purity,the quality inspection method of Isatis indigotica seeds were studied and the best inspection conditions for each index are determined.The results showed that quartering method for sample sampling,determination of weight by a thousand particle method,and the filter paper was used as a germination bed and the germination rate was measured at 15?.The first time of the seed germination record was the 6th day and the final time was the 10th day.The moisture content of seeds was tested with stoving for 6 h at105±2?.The testing for the seed vigor by dyeing for 1 h with 0.5%TTC under 30?showed the best.The protein was extracted by mercapto ethanol and the seeds purity was determined according to the similarities and differences of the seeds protein bands.2.50 samples of Isatis indigotica seeds and fruits collected from different place as the research object.According to the 1000-fruit weight,germination rate,moisture content and clarity index,Isatis indigotica fruits were divided into 3 levels.Respectively,the first grade were at least 97%in fruit clarity,at least 8.6 g in 1000-fruit weight,86%in fruit germination rate,at most 9.0%in fruit moisture content;the second grade seeds were at least 93%in fruit clarity,at least 7.7 g in 1000-fruit weight,74%in fruit germination rate,at most 9.8%in fruit moisture content;the third grade seeds were at least 89%in fruit clarity,at least 6.5 g in1000-fruit weight,55%in fruit germination rate,at most 10.6%in fruit moisture content.According to the viability,1000-seed weight,germination rate,moisture content and clarity index,Isatis indigotica seeds were divided into 3 levels.Respectively,the first grade were at least 98%in seed clarity,at least 95%in seed viability,at least 3.2 g in 1000-seed weight,95%in seed germination rate,at most 9%in seed moisture content;the second grade seeds were at least 95%in seed clarity,at least 85%in seed viability,at least 2.9 g in 1000-seed weight,92%in seed germination rate,at most 10%in seed moisture content;the third grade seeds were at least 92%in seed clarity,at least 75%in seed viability,at least 2.6 g in1000-seed weight,88%in seed germination rate,at most 11%in seed moisture content.3.The Isatidis Radix was determined by root length,root diameter,scab level,branch root,root weight,moisture,ash,acid insoluble ash,extract,?R,S?-epigoitrin,Indigo and Indirubin.Then the Isatidis Radix was divided into three grades based on root diameter and extract,i.e.the first grade were at least 8.5 mm in root diameter,at least 45%in extract;the second grade were 5.88.5 mm in root diameter,4345%mm in extract;the third grade were3.55.8 mm in root diameter,2545%in extract.4.The contents of?R,S?-epigoitrin,Indigo and Indirubin of Isatidis Radix in Gansu?S1?,Henan?S2?,Heilongjiang?S3?and Xinjiang?S4?was determined by high performance liquid chromatography?HPLC?and the fingerprints of 4 kinds of Isatis Radix were established.The results showed that the content of?R,S?-epigoitrin was S2>S1>S3>S4,the content of Indigo and Indirubin was S2>S4>S1>S3.The fingerprint results showed that the similarity between the four medicinal herbs fingerprints with the control fingerprint was S4>S3>S1>S2.It indicates that the fingerprint of medicinal materials in Xinjiang was closest to the control,while the fingerprint of Henan medicinal materials was quite different from the control. |
