In this paper,the genetic diversity index,population genetic structure and genetic differentiation of two populations for Hooded Crane(Grus monacha)and Red-crowned Crane(Grus japonensis)were studied using 11 universal SSR loci.The aim of the study was to analyze the factors that affect the genetic diversity of the two populations and explore the causes of the Population genetic structure.It's to provide scientific basis for the artificial breeding and management of these two populations,On the other hand,to provide a scientific protection and management advice to ensure the healthy and stable development of the two populations.The main results are as follows:1.Genetic diversity and population genetic structure of Hooded CraneThis experiment take 37 Hooded Crane samples in five zoos by 11 SSR loci were amplified and successfully amplified stable results.The results showed that there are 96 alleles were amplified,the average allele was 8.7,the rare allelethe was 36,average effective allele was 5.4.The average observe heterozygosity was 0.5849;The average expected heterozygosity was 0.7858.The average polymorphism information content was 0.7379,there are 10 highly polymorphic loci and 1 moderately polymorphic loci.Genetic diversity is at a high level.HardyWeinberg equiliberum(HWE)test showed that 3 SSR loci were not in HWE.Fis in-0.0661 ~ 1,the average value is 0.2065,the results showed that there was a phenomenon of homozygous excess of heterozygote deficiency in hooded crane population.2.Genetic diversity and genetic relationship of Red-crowned CraneThis experiment take 58 red-crowned crane samples in Shenyang Forest Zoo,including 18 pairs of matching breeder,9 juvenile cranes and 13 a few years ago breeding red-crowned cranes.The results showed that there are 68 alleles were amplified,the average allele was 6.2,the rare allelethe was 24,average effective allele was 3.1.The average observe heterozygosity was 0.5892;The average expected heterozygosity was 0.5866.The average polymorphism information content was 0.5533,there are 7 highly polymorphic loci and 4 moderately polymorphic loci.Genetic diversity is at a high level.Hardy-Weinberg equiliberum(HWE)test showed that 2 SSR loci were not in HWE.There were close relatives mating in the population.8 SSR loci were selected for paternity test,the cumulative non-parental exclusion rate was 0.9314,the identification rate was 100%,and the results could be applied to the redcrowned crane paternity test.
