Triploid Populus simonii×P.nigra Hybrid,because of its rapid growth,high resistance is widely used in the Northeast region.Many physicists have revealed the physiological and molecular mechanisms of Triploid Populus simonii×P.nigra Hybrid under salt stress.In this study,Triploid Populus simonii×P.nigra Hybrid were obtained by hybridization.Cloning showed obvious rapid growth,but the ability to resist salt and has not been analyzed.Triploid Populus simonii×P.nigra Hybrid clones were used as materials for growth traits.The physiological indexes and proteomics were determined under salt stress,and the following results were obtained : 1.Analysis of variation of Growth Index of Populus simonii×P.nigra Hybrid under different salt stressThree clones of XY4.1,XY3.2 and XY as materials,and the chl,the leaf relative water conten,leaf fresh weight,dry weight of leaves,stems water content,dry weight of stem and stem fresh weight were studied under salt stress.The results showed that the chlorophyll content,leaf water content,leaf fresh weight,leaf dry weight,stem water content,stem fresh weight and stem dry weight of different clones were decreased with the increase of salt concentration,and with the increase of salt concentration.the salt stress concentration of 75mmol/L was found to be the most suitable salt stress concentration for clones.2.Analysis of physiological index variation of Triploid Populus simonii×P.nigra Hybrid(XY3.2)under salt stressThe results showed that 10 physiological indexes(conductivity,POD,SOD,CAT,soluble sugar,proline,malondialdehyde,MDA,reactive oxygen species and plasma membrane H +-ATPase)were measured under the salt stress.These physiological indexes showed a downward trend after an initial increase.POD,SOD and H +-ATPase were all increased before 12 days of stress,and decreased after 12 days.The CAT,soluble sugar,proline,MDA,soluble protein and reactive oxygen species were 8 days before salt stress,and decreased after 8 days.3.Effects of Triploid Populus simonii×P.nigra Hybrid(XY3.2)leaf chlorophyll fluorescence parameters under salt stressThe clones of XY3.2 as materials,the chlorophyll fluorescence parameters results show,Fm,?PS? and ETR showed a decreasing trend under salt stress,and the chlorophyll fluorescence parameters were measured under salt stress.While NPQ and qN showed a gradual increase trend,the qP and Fv/Fm showed a unimodal trend.The results showed that could inhibit the original light energy conversion efficiency and the activity of PS?,increase the dissipation of PS? non-radiation energy,and the defense ability of photosynthetic system under salt stress was enhanced by clone XY3.2.4.Differential protein expression analysis in Triploid Populus simonii×P.nigra Hybrid(XY3.2)under salt stressThe differentially expressed proteins were analyzed under the salt stress,and differentially expressed proteins of 86 were identified.The number of different proteins was different in different time,and the difference was the most significant 32 of them.Application of GO function annotation software analysis of the above protein function,specifically divided into energy metabolism,redox,photosynthesis,signal transduction,transport protein,resistance protein,biosynthesis and metabolism and unknown protein 10 categories: energy metabolism accounted for 19 %,photosynthesis accounted for 6.9%,photosynthesis accounted for 26%,signal transduction accounted for 2%,transport protein accounted for 3%,resistant protein accounted for 8%,biosynthesis and metabolism accounted for 6% and unknown protein accounted for 23%.From the above analysis,it was explained that the XY3.2 was the most affected by photosynthesis and energy metabolism,followed by redox reaction.To sum up,first determine the most sensitive salt stress concentration of 75 mmol / L..The chlorophyll content,enzyme activity and fluorescence characteristics were determined by 75 mmol/L NaCl treatment of Triploid Populus simonii×P.nigra Hybrid(XY3.2),and the suitable number of days was determined.Finally,the differential expression of leaf protein was determined and the protein expression of Triploid Populus simonii×P.nigra Hybrid(XY3.2)was analyzed under salt stress.The molecular mechanism of Triploid Populus simonii×P.nigra Hybrid was studied and the data of protein level were provided.And the mechanism of Triploid Populus simonii×P.nigra Hybrid salt tolerance mechanisms also provide targets for salt polyploid breeding clones.
