Anhui province is the national important production area of green tea,which owns lots of local high quality tea and excellent local species population.After a preliminary screening,10 local excellent species population were choosen and investigated.We then went to the relative core areas,and recorded the geographical coordinates and altitude of different species.Morphology characters and main components of local excellent species population which were suitable for brand tea were analysized.The genetic relationship of these 10 different areas was build and anylized by using SSR method,and phylogenetic tree was also established.Tea DNA extraction method was optimized according to the needs.The results are as follows:1.We did field investigation of 10 selected local famous quality green tea groups in Anhui province,and recorded the distribution and collected the growth area image data of corresponding groups.2.We observed the individual morphological indexes of typical species,and summarized the morphology characters of local species population in same areas and different areas.3.We measured the catechins and caffeine as well as total amino acid content of selected samples,compared these chemical composition content and worked out the average content by using statistical analysis technique,which samples were came from the same and different areas.4.Since SSR requires large amount of high quality DNA genome,we improved the extraction method systematically,optimized the formula of loading buffer,and changed the extraction steps.Compared with the traditional methods,the extraction time was shortened drastically,and the quantity and quality of DNA genome were increased,and the cost was also reduced.5.We used 50 pairs of SSR primers metioned in published papers to analysize the genetic diversity of 65 tea samples from 10 areas,and detected 224 alleles and 392 genotypes.Each primer could detect 2-12 alleles and 2-16 genotypes,with the average number was 4.98 and 7.84,respectively.(Nei)was between 0.0465 and 0.7628 when average number was 0.4751 and the PIC was 0.4588.(He)was between 0.1000(G120)and 0.773(G112),when average number was 0.4826.(Ho)was between 0.0476(G180)and 0.5238(G127)with the average number of 0.2686.(I)was between 0.1023(YG3)and 1.5363(G112),when average number was 0.7869.These suggested the genetic diversity was abundant.The phylogenetic tree was build by using NTSYSpc-2.10 software.
