Paris polyphylla Smith var.chinensis(Franch)Hara.(Paris polyphylla var.chinensis)has become a treasured and endangered Chinese herbal medicine in China.Because of its scarcity of wild resources and large demand,the price of this medicine have jump from 100yuan/kg to 1000 yuan/kg in ten years.The artificial cultivation and rapid propagation of Paris polyphylla var.chinensis are imminent.Since 2012,the author successively took part in two related topics of replanting,and the project team conducted a series of studies on the key technologies for rapid breeding and standardized planting of Paris polyphylla var.chinensis.We found that the Paris polyphylla var.chinensis had slow growth,low vegetative reproduction and reproductive coefficient,high cost,low germination germination rate,slow germination,high tissue culture cost,and low survival rate.Studying the seed germination rate and activity is an important road for sustainable development and large-scale planting of Paris polyphylla var.chinensis resources.Considering the adaptability of the varieties,were choice the seeds of Paris polyphylla Smith var.chinensis as research materials.By measuring seed vigor after treatment with different methods,we studied the effect of these treatments on seed vigor.Through three methods,we soakes the no-deseeded seeds and deseeded seed with fresh water in differet periods of time;Deal with different seeds with different concentrations of potassium permanganate for 48 h;Drying in an oven at 105°C,and messured the water content every two hours,and 5 groups of water content were obtained after 8 h.The activities of POD,SOD and CAT,and the content of MDA were determined after treatment with the above different methods.The experimental results are as follows:1.The effects of fresh water soaking on seed vigor at different times: The viability of Paris polyphylla var.chinensis’ s seed not to be seeded and the seed to be seeded was increased first and then decreased as the soaking time prolonged.The maximum value was reached when the seed coat was soaked for about 2 days.The seed viability of the seedless seedling reached its maximum value in the fresh water soaking for 5 days;This result has a strong guiding significance for the seed treatment of Paris polyphylla var.chinensis before sowing.2.The effects on seed vigor of potassium permanganate soaking with different concentrations: The viability of seed not yet deseeded increased first and then decreased as the concentration of potassium permanganate increased,the vigor was always reduced,Seed coat seeds soaked with 0.10 g/L potassium permanganate for 24~48 h to increase their vigor.And unseeded seed soaked with 0.50 g/L potassium permanganate for 48 h to increase their vigor.Paris polyphylla var.chinensis’ s seed treatment before sowing has a certain guiding significance.3.Seed vigor: The seed vigor of Paris polyphylla var.chinensis decreased with the decrease of water content,and its vitality was the highest when the water content was more than 60%,When the water content of the seed is less than 30%,the vitality is significantly reduced.Reducing the moisture content is not conducive to keeping the vitality of seeds of Paris polyphylla var.chinensis.This result has certain guiding significance for the processing and storage of seeds after harvest.Through the above studies and the related literature,Initially drafted Paris polyphylla var.chinensis Seed Quality Standard and its Inspection Technical Regulations(SOP).
