Peste des petits ruminants(PPR),as a violent and contagious disease,caused by Peste des petits ruminants virus(PPRV)and characterized by a high fever and pneumonia.PPRV is an enveloped virus,with a single-strand,negative-sense RNA genome,which is belong to Paramyxoviriade Morbollivirus member.As a conserved molecular protein chaperone in cells,HSP70 can provide important assistance for proper folding during protein synthesis.Heat shock proteins will be secreted extensively to protect organisms from damage when they are stimulated by heat stress,glucose starvation or virus infection.According to different molecular weights,heat shock proteins can be divided into small molecular weight HSP family,HSP60 family,HSP70 family and HSP90 family.At present,people have conducted extensive research on relation between heat shock protein 70 and virus,but few studies had investigated in the heat shock protein after animals were infected with PPRV.In this study,goat fetal fibroblasts(GFF)were infected with PPRV which was to study the expression of HSP and its effect on PPRV proliferation,thus providing a theoretical basis and scientific basis for studying the pathogenic mechanism of this virus.The results are as follows:1.The expression change of HSP after infection.The expression of HSP70 was detected by Real time-PCR and Western blot at different times after infection.The results showed that the infection of PPRV significantly increased the expression of HSP70 both in mRNA and protein level,consistent with the pattern of virus proliferation;HSP32 and HSP90 have not the same changes.In addition,Vero cells got a different conclusion,indicating that the host cells and the model cells have different mechanisms.2.Results of viral proliferation treated with Lentivirus.We constructed pCDHCMV-HSP70 in the present study.Western blot analysis showed increase the expressio n level of HSP70 can promote the viral proliferation in GFF cells.There is a positive correlation between the HSP70 and virus.3.Results of viral proliferation after down-regulated HSP70.In this study,we used specific inhibitors of HSP70 proteins,quercetin and lentivirus packaging method reducing the protein level of HSP70,the results both showed that positive correlation between HSP70 expression level and PPRV replication in the cells.In other words,HSP70 protein plays a significant role in the proliferation of PPRV.In summary,the present study shows that PPRV infection can up-regulate the exp ression of HSP70 in GFF,and the regulation of HSP70 expression can significantly af fect the proliferation of PPRV in cells,and they are positively related.These results p rovide important reference to the targets of anti-PPRV drugs,and also provide new id eas for the prevention and control of virus.
