Genetic Diversity Based On SSR And Its Association Analasis With Phenotypic Traits In Armeniaca Vulgaris Var.ansu

This paper took 60 clones of Armeniaca vulgaris var.ansu which came from Armeniaca sibirica National Forest Germplasm Resource Preservation Repository as research object.18 quality traits were observed and 25 quantitative traits were measured.And we used 120 pairs of primers to amplify SSR markers.Then genetic diversity of phenotype traits and SSR markers were analyzed.And on this basis,the correlation analyses between phenotypic traits and SSR markers were carried out.The summarized results were as follows.(1)The leaf shape,petal color,pulp taste,almond taste and other main traits among 18 quality traits were consistent with phenotypic characteristics.The variation coefficients of 25 quantitative traits were between 7.8% and 42.6%,and the variation coefficients of 22 quantitative traits were more than 10%.(2)The cluster results mainly reflected the difference of phenotypic traits.In the first category,the index of leaf shape was the smallest,and the phenotypic traits were mediocre.The second category not only had the largest value of fruit,nutlet and kernel characters but also had the largest growth of shoot.In the third category,the index of leaf shape was the largest,kernel rate was the smallest,and there were many variation types of flower and fruit.In the fourth category,fruit shape index,nutlet shape index and kernel shape index were the largest,and nutlet rate and kernel rate were larger.In the fifth category,nutlet rate and kernel rate were largest,and there were many variations of flower.Each category respectively had different applications of the fruit,nutlet,kernel,ornamental and the special type.(3)64 pairs of primers with high polymorphism were screened out of 120 pairs of primers.The average number of alleles was 7.438.The average number of effective alleles was 3.359.The average polymorphic information content was 0.639.Clones were divided into three subgroups by STRUCTURE,and the results showed the genetic background information of different clones and the genetic diversity of provenance and germplasm characteristics.Meanwhile clones were grouped into 4 groups by NTsys,and the results of clustering mainly reflected the differences in provenance and germplasm characteristics of individuals(4)There was no significant correlation between cluster of phenotypic traits and the two results of SSR markers clustering.But there was significant correlation with two molecular softwares.(5)The results of association analysis between phenotypic traits and SSR markers showed that there was extensive linkage disequilibrium among SSR loci.178 loci were paired in the case of p<0.01 and 142 loci were paired in the case of D??0.5.Under extremely significant conditions(p<0.001),14 loci and 13 loci were respectively associated with 10 traits through using two models of GLM and MLM.After integrating the mutual association markers,it showed that 12 loci respectively associated with 9 traits.To conclude,the results of this study showed that the genetic diversity of Armeniaca vulgaris var.ansu based on phenotypic traits and SSR molecular markers was rich.The clustering results reflected the differences in the characteristics of provenances and germplasm,and 12 SSR loci related to 9 performance traits were obtained by SSR molecular markers association analysis,and the results was to provide theoretical guide and technological support for the identification,evaluation,utilization and breeding research of germplasm resources in Armeniaca vulgaris var.ansu.
