Reasearch On Celastrus Orbiculatus Extractin Treatment Of Liver Fibrosis In Rats

Purpose:To investigate the therapy effection of C. orbiculatus ethyl acetate extracts (COE) in ClCl4-inducing liver fibrosisin Sprague-Dawley rats, and make a discusion of the undelying mechanisms.Method60 SD rats were randomly divided into Normal group, Control group and Experimental group. The Normal group was no any treatment, The Control group was in trap-eritoneal injection with saline (0.75 mL/kg), Experimental group was in traperitoneal injection with 15%-carbon tetrachloride (0.75 mL/kg), three time every week.8 weeks later, The rats were randomly executed, the changes of livers pathology were observed by light microscope, these pathologic changes demonstate a succesfull modeling. Then the experimental group were divided into model group?COE groups(low, medium, high) and Tanshinone group, each group had 10 rats.Then the model group was intragastric administration with saline. The concentration of COE was setted by 12.5,25,50 mg/kg. COE was intragastric administration for 20 days. Tanshinonegroup was intraperitoneal injection with Tanshinone?A Sulfonic Sodium (20 mg/kg) for 3 days. After treatment, serum index was detected by ELISA, includeal bumin (ALB), glutamic-pyruvic transaminase (ALT), procollagen ? (PC?), type iv collagen (CIV), transformating growthfactor-?1 (TNF-?) and tumour necrosises factor-? (TNF-?). Pathol-ogic changes of liver were observed by hematoxylin-eosin staining.ResultThe liver tissue of the Experience group rats were observed underly microscope that hepatic lobule strucure destroyed,a numerber of connective tissue proliferation discorvered aroud portal area and central vei-ns, hepatocyte changes showed edema, degeneration and focal cytolysisnecrosis, which demostrated a succussfull modeling was biudling. The result of Liver function index was that serum ALB and ALT had no markedly change in COE low-group,compared with model group (P>0.05). But in COE medium,high-groups and Tanshinone group,sersum ALB showed increased and sersum ALT decreased (P<0.05). compared with model group, the express of serum PCIII, CIV, HA, LN, TGF-?1 and TNF-? had no diffrence in COE low-group (P>0.05), but markdely decresed in COE medium, high-groups and Tanshinone group (P<0.05). The results of HE:compared with model group, COE low-group had no influence, the fibrosis degree had significantly improved in COE medium, high-groups and Tanshinone group.ConclusionCOE effectiently improved quality of life and liver function, releaved degree of fibrosis of clcl4-inducing liver fibrosis in rats. Its mecheism was possiblly associated with reducing epresstiong of TGF-?1and TNF-?.
