Objective RNAi technology to suppress the key gene REV3 L expression that is associated with the translesion DNA Synthesis, combined with anticancer drugs on the influence colon carcinoma sw480 cell xenografts in nude mice through lastingly inhibition effect on REV3 L,to investigate the mechanism of to the growth of implant tumor and REV3 L function.Methods Athymic mice were subcutaneously transplanted with human colon careinoma SW480 cells to establish xenografts model. By RNAi technology, REV3 L expression was suppressed in SW480 cells, single and joint of sensitive cancer chemotherapy drugs oxaliplatin and the traditional Chinese medicine sophoridine were used to explore the inhibition on REV3 L expression, we analysed the expressed by performing immunohistochemistry analysis and real-time fluorescence quantitative PCR. By HE staining, TUNEL techniques relevant organizations morphological indicators, statistical methods(such as: the weight of nude mice, tumor volume changes in cell morphology and intensity, tissue cell apoptosis status) detection growth-inhibition of xenografts before and after the anticancer drugs treatment, and inhibition REV3 L gene expression in the growth of xenografts of nude mice.Results Interior group comparison :the detection of indicators were not significant statistically between the two control groups(blank and negative); There is significant statistically among the treatment groups(REV3L gene low expression + single and combination therapy) and two control groups(blank and negative single joint medication),P <0.05. There is not statistically significant between oxaliplatin and sophoridine.Conclusion By RNAi technology, expression of REV3 L gene was suppressed in colon cancer cells xenografts in nude mice; the growth of xenografts was lower supplementing by a single agent(the oxaliplatin and sophoridine). It can inhibite strongly the xenograft growth, proliferation by the combination therapy(oxaliplatin + sophoridine to). The effect of Anticancer Drugs was Evaluated by anticancer drugs therapy and inhibition effect of the REV3 L gene involved in DNA damage. There treatment are used as a new attempt of cancer treatment interventions, and also will help us to further understand the incidence of colon cancer development and prognosis for cancer prevention, diagnosis and treatment of the theoretical basis and clinical guiding.
