Objective To obtain and investigate platelet micropaticles (PMP) in manually prepared condensed platelet. Methods The content of PMP in platelet concentrates stored at day 1, 3,5,7,9,11 was detected by flow cytometry. Platelet concentrates stored at day 1,8,15, 22,29 were centrifugated, isolated platelets were used to analyse the effect of centrifuge shear force by the detected CD62p and phosphatidylserine, while the supernatant was used to obtain PMP. Related physical, chemical and physiological function of obtained PMP was analysed by following research:size of PMP particles, surface protein receptor and phosphatidylserine, thrombin generation ability. Platelet activation and apoptosis indicators in platelet concentrates stored at day 0,1,3,5,7,9,11 were detected by flow cytometry, CD62p was used as platelet activation indicator, ?(?)m and phosphatidylserine were used as platelet apoptosis indicator. The source of PMP was analysed by the detected platelet physiological state. Results The content of PMP was significantly increased in platelet concentrates when stored at day 9, and centrifuge shear force has no effect on the obtention of PMP. The obtained heterogeneity PMP retain GP I b(CD42b), GP ?b-?a(CD61), P-selection(CD62p) and phosphatidylserine on the surface. PMP can also promote thrombin generation in vito. The detection of platelet activation and apoptosis indicators indicate that platelets were highly activate and apoptosis after platelet concentrates stored 5 days. Conclusion PMP was significantly released from platelet concentrates when stored 9 days, and PMP can be obtained directly by dual centrifugal. PMP remains GP I b, GP II b-?a, P-selection and PS on the surface, and have hemostatic function in vito. The hemostatic PMP were released from highly activation and apoptosis platelet.
