| In the past ten years,cucurbit[n]uril chemistry advances in the blossoming field because of their exceptional recognition properties in aqueous medium.They have also become key units in various mole cular assemblies,sensors,drug carrier,as well as in advanced material-s.The scope of this article is limited to the main family of cucurbit[n]urils(n=5,6,7,8,10).In particular, the cucurbit[7]uril(CB[7]) host has received considerable recent attention because of its solubility in aqueous solution.We use fluorescence spectroscopy,UV-vis spectroscopy,Nuclear magnetic resonance spectroscopy,density functional theory calculation to study the interaction between drug and cucurbit[7]-uril molecules,using berberine and cucurbit[7]uril, palmatine and cucurbit[7]uril,coptisine and cucurbit[7]uril as fluorescent probes to study cucurbit[7]uril and guest molecules recognition.This paper mainly divided into the following four aspects:1. Firstly,introduces the history of supramolecular, by order of the development of supramolecular respectively introduces the crown ethers,cyclodextrins,calixarene and cucurbit[n]uril, then introduce the research background and significance of the supramolecular, especially introduces the research significance of cucurbit[n]uril, on the basis of predecessors exploration our team has done in the research of the synthesis cucurbit[n]uril(n=5,6,7,8), and step by step into purified to obtain the host of cucurbit[7]uril in our experiment,research progress in supramolecular spectrometry, our laboratory has carried on the related research, mainly divided into fluorescence sensitization method and fluorescence quenching method.Finally,introduced the thesis research in this paper.2. Camphor(1,7,7-trimethyl-bicyclo[2.2.1]hepta-2-one),it has no fluorescence,we use three kinds of fluorescent probes: the cucurbit[7]uril and berberine hydrochloride(CB[7]-BER), cucurbit[7]uril and palmat Ine(CB[7]-PAL),cucurbit[7]uril and coptisine(CB[7]-COP),and by means of fluorescence spectrometry,UV-vis spectroscopy and Nuclear magnetic spectroscopy and Molecular model of theoretical calculation methods to study the cucurbit[7] uril and camphor molecular interaction,camphor molecules and cucurbit[7]uril form a stable 1:1 inclusion complex,and through the nonlinear quadratic fitting, the combination constants of camphor molecule and cucurbit[7]uril(0.92±0.02)x107M-1,(1.4±0.02)x107M-1,(1.51±0.02)x107M-1and for coptisine, berberine hydrochloride, palmatine and cucurbit[7]uril, the combination constant for KCB[7]-COP=1.28x104M-1, KCB[7]-BER=1.77x105M-1, KCB[7]-PAL= 1.1x105M-1,it shows the binding force of camphor and cucurbit[7]uril is greater than the fluorescence probes and cucurbit[7]uril molecules, so camphor can make the probe molecules leave from the cavity of the cucurbit- [7]uril.And we can use it in actual samples,we achieved satisfactory results in the detection,we are studying more practical sample testing.3.In experiment of cucurbit[7]uril and gold nanoparticle assembly, first,gold nanoparticles were synthesized and characterized by the transmission electron microscopy(SEM) and UV-vis-spectrometry,it is concluded that the diameter of the gold nanoparticles was 12 nm, then we used Chromatography for the purification of horseradish peroxidase(HRP) to get a pure substance.Under certain conditions we make conjugate of Ab2-Au NP-HRP and Ab2-HRP,the products will be tested with Zeta potential meter,the results show that potential is negative,we concluded that horseradish peroxidase has been attached on the surface of gold nanoparticles,the products were added into the solution of cucurbit[7]uril,respectively.The great changes have been taken place in the solution of the fluorescence intensity,and the combination has the function of signal amplification, Ab2-Au NP-HRP signal is Ab2-HRP 5 times, indicating the high sensitivity of the method,it can be used as a sensor to actual testing, related research is in progress.4.Chelerythrine is a kind of isoquinoline alkaloid, it can be used in antimicrobial, antibacterial and anti-inflammatory, we will study this matter because of its use.First of all, from the material of the fluorescence properties to study, itself has no fluorescence,so it is difficulty to use fluorescence technique, However,we used a supramolecular cucurbit[7]uril as the carrier, studies the interaction between them,one of the most interesting thing is that we found no fluorescent chelerythrine into the cucurbit[7]uril solution make the fluorescence intensity increased obviously,which makes us find no fluorescent signal substance can be detected by fluorescence spectrum method.With the change of p H,there is a change in fluorescence spectra,UV-vis spectra,it may be the structure of this kind of material along with the change of p H changes.This phenomenon can be used in bio-sensing,we can pass changes in the p H of the environment around,it has great significance in the drug release, and we conducted molecular simulations, calculating the best ratio of combination of them,sensor applications are under study. |
