Objective: By intervening the myocardial infarction rat model with Zhi-Long Huoxue Tongyu capsule,than make rat cardiac ultrasound,observed in the rat Serum levels of b-type natriuretic peptide(BNP)levels,and the myocardial local angiotensin II(ANG II),angiotensin II receptor 1(AT1R),and transforming growth factor beta 1(TGF-?1)protein content after 8 weeks.To explore the mechanism of Zhi-Long Huoxue Tongyu capsule to improve myocardial infarction in the rat myocardial remodeling.Methods: By ligature left anterior descending coronary artery ligation rats copy the myocardial infarction model in rats.Choose 70 healthy rats,were randomly divided into control group and 10 and surgical group 60,after the success of the surgery group building randomly selected from 50,were randomly divided into model group,the Captopril group,Zhi-Long Huoxue Tongyu capsule low-dose group,Zhi-Long Huoxue Tongyu capsule dosage group,Zhi-Long Huoxue Tongyu capsule high-dose group,10 rats in each group.In control group and model group normal saline lavage daily,the rest respectively of the corresponding drug lavage between groups,,keep 8 weeks.Observed in the rat cardiac uhrasonography,Serum levels of b-type natriuretic peptide(BNP)levels,And the myocardial local angiotensin II and angiotensin II receptor 1,transforming growth factor beta 1 protein content after 8 weeks.and return the heart tissue for observation HE dyeing lines of infarction area.Results:(1)The rats cardiac uhrasonograph: After 8 weeks,The left ventricular end-diastolic dimension(LVEDD)and left ventricular end-systolic dimension(LVESD)of model group were rise obviously,the left ventricular ejection fraction(LVEF)?left ventricular fractional shortening(LVFS)were drop obviously,compared with the control group,the difference was statistically significant(P<0.05),Compared with the model group,the LVESD reduce slightly and LVEF?LVFS increase slightly of Zhi-Long Huoxue Tongyu capaule dosage group(P<0.05);The LVEDD ? LVESD reduce obviously and the LVEF?lLVFS increase obviously of Zhi-Long Huoxue Tongyu capaule high-dose group(P<0.05),it has a similar results with the Captopril group(P>0.05);(2)Serum levels of BNP levels: After 8 weeks,compared with the control group,the serum levels of BNP levels increase obviously of the model group(P<0.05),compared with the model group,the BNP of Zhi-Long Huoxue Tongyu capaule low-dose group reduce slightly(P<0.05),the BNP of Zhi-Long Huoxue Tongyu capaule dosage group reduce slightly(P<0.05),the BNP of Zhi-Long Huoxue Tongyu capaule high-dose group reduce obviously(P<0.05),it has a similar results with the Captopril group(P>0.05);(3)The myocardial local ANG II and AT1 R protein content: After 8 weeks,compared with the control group,the myocardial local ANG II and AT1 R protein content increase obviously of the model group(P<0.05),compared with the model group,the the myocardial local ANG II and AT1 R protein content of Zhi-Long Huoxue Tongyu capsule low-dose group reduce slightly(P<0.05),the myocardial local ANG II and AT1 R protein content of Zhi-Long Huoxue Tongyu capsule dosage group reduce slightly(P<0.05),the myocardial local ANG II and AT1 R protein content of Zhi-Long Huoxue Tongyu capsule high-dose group reduce obviously(P<0.05),it has a similar results with the Captopril group(P>0.05);(4)The myocardial local TGF-?1 protein content: After 8 weeks,compared with the control group,the myocardial local TGF-?1 protein content increase obviously of the model group(P<0.05),compared with the model group,the myocardial local TGF-?1 protein content of Zhi-Long Huoxue Tongyu capsule low-dose group has a similar results with it(P>0.05),the myocardial local TGF-?1 protein content of Zhi-Long Huoxue Tongyu capsule dosage group reduce slightly(P<0.05),the myocardial local TGF-?1 protein content of Zhi-Long Huoxue Tongyu capsule high-dose group reduce obviously(P<0.05),it has a similar results with the Captopril group(P>0.05);(5)Observation HE dyeing lines of infarction area: In the control group,ocardial cell staining balance,neat horizontal stripes,cell morphological structure integrity;In the model group,cardiac muscle fibers have thickeninged,fractured,arranged in disorder,Inflammatory cells infiltration,Myocardial fibrosis were very seriously;There is a small amount of cardiomyocyte hypertrophy in Zhi-Long Huoxue Tongyu capsule high-dose group,dosage group,low-dose group and the Captopril group,myocardial fibrosis is lighter than model group,The uninfarction area neat myocardial cells,the inflammatory cells infiltration is less than Model group,the cardiac fibrosis about Zhi-Long Huoxue Tongyu capsule high-dose group and the Captopril group are decreased significantly than Zhi-Long Huoxue Tongyu capsule dosage group and low-dose group.Zhi-Long Huoxue Tongyu capsule can reduce myocardial infarction rats LVDD,LVSD,and improve myocardial infarction rats LVEF,LVFS,improving myocardial structure,lower serum BNP levels in rats after myocardial infarction,improve heart function.Zhi-Long Huoxue Tongyu capsule can inhibit myocardial local Ang II,AT1 R,TGF-beta 1 protein expression,inhibition of myocardial extracellular matrix formation and deposition,inhibiting myocardial fiber cell hyperplasia and alleviate myocardial fibers,inhibit ventricular remodeling...
