Objective:Some pro-wound healing factors change in the quantity and quality during diabetic foot ulcer(DFU)wound healing.Changes in the production of growth factors and cytokines have negative effects on wound healing in diabetic patients.Angiomotin binds to angiostatin and plays a vital role in angiogenesis,promoting tubule formation and growth factor-induced migration of endothelial cells.Ischemia-reperfusion(I/R)injury can be a factor in the formation of chronic skin wounds.We determined respectively Angiomotin protein expression in skeletal muscle sampled from diabetic rats with hind-limb ischemia and wound granulation tissue sampled from diabetic rats with I/R skin ulcers by Western blotting to investigate the regulatory mechanisms of the Angiomotin on angiogenesis in diabetic ischemic ulcer-healing and to analyze potential relations between Angiomotin and I/R injury,so as to further research the etiological mechanisms of chronic refractory foot ulcer.Methods:1.Part one:Forty-eight Sprague-Dawley(SD)rats were divided into four groups according to their blood glucose and blood supply.A rat diabetic hind limb ischemia model was created by excising the femoral artery and all its major branches.Western blotting was used to determine the Angiomotin expression in skeletal muscle.2.Part two:48 SD rats were divided into four groups according to their blood glucose and I/R cycle protocol.I/R injury was induced by applying and removing a permanent magnet to a femoral region of rat skin under which a ferromagnetic steel plate was implanted.The Angiomotin expression in wound granulation tissue was also determined by Western blotting.Results:1.Part one:The Angiomotin was expressed in skeletal muscle from diabetic rats with hind-limb ischemia.Expression of p130-Angiomotin in the group A was the lowest among the four groups(P =0.01),expression of p80-Angiomotin,however,was no significant difference.2.Part two:The Angiomotin was expressed wound granulation tissues from diabetic rats with I/R skin ulcers.Expression of p80-Angiomotin in the group B was the lowest among the four groups(P =0.00),nevertheless the differences of p130-Angiomotin were not statistical significance.Conclusion:1.Expression of Angiomotin is decreased by diabetes and hind-limb ischemia.Further analysis showed that compared with diabetes,hind-limb ischemia is more impact on expression of p130-Angiomotin.2.Expression of Angiomotin is decreased by I/R injury.
