Objective:1.To compare whether there were defferences of expression of Amot and p80/p130Amot in granulation tissue between patients with diabetic foot ulcer and non-diabetic patients;2.To observe whether hyperglycemia and peripheral vessel disease had an influence on the expression of Amot in granulation tissue of diabetic foot ulcer patients.2.After 6-months' standard treatment,to analyze the relation between Amot as well as p80/p130Amot and diabetic foot ulcer healing through Logistic regression model.Methods:52 patients with diabetic foot ulcers who wereb hospitalized in the foot department of the Metabolic Disease Hospital of TianJin Medical University from September,2009 to June,2010 and 6 patients without diabetes as control group were recruited.According to GHbA1c and ABI,52 patients with diabetic foot ulcer were divided into hyperglycemia group(A group),non-hyperglycemia group(B group),ischemia and hyperglycemia group(C group)and ischemia and non-hyperglycemia group(D group)We got granulation tissue of foot ulcers during debridement or surgery and measured the expression level of Amot in granulation tissue through Western blotting.Then we compared whether there were differences for expression level of Amot and p80/p130Amot among 5 groups of patients and observed whether hyperglycemia and peripheral vessel disease had influence on expression level of Amot and p80/p130Amot for 52 patients with diabetic foot ulcers.Collecting the general data,clinical data and biochemical data of 52 patients with diabetic foot ulcer after hospitalization,according to whether foot ulcers healed or not,52 patients were divided into healing group and unhealing group after 6-months' standard treatment.We explored the relation between Amot,p80/p130Amot and diabetic foot ulcers healing through univariate analysis;Healing or not of foot ulcers as dependent variant,we brought the risk factors above into Binary Logistic Regression Model and analysed the independent risk factors of healing of diabetic foot ulcers.Results:Through Western Blotting measurement,we found that 4 groups of patients with diabetic foot ulcers and patients without diabetes had the expression of p-80 and p-130Amot in granulation tissue and p-80Amot was dominant in 5 groups of patients.expression level of amot in 4 groups of patients were lower than that of control group,there was a significant difference between 5 groups(p=0.000).C group had the lowest expression level of Amot in 4 groups of patients with diabetic foot ulcers,the expression of Amot of C group was lower than A group,there was significant difference between C and A group(p=0.000).But there was not significant difference between C and D group(p=0.052).Comparing the expression level of Amot of A group in hospitalization with that of A group after intensive control of blood glucose of 2 months,there was not significant difference(p=0.232).52 patients with diabetic foot ulcers,34 healed,Univariate analysis indicated that wound area,expression level of p80Amot,p80/p130Amot and peripheral vessel disease were the risk factors of ulcer healing.Binary Logistic regression Model demonstrated that peripheral vessel disease was the independent risk factors of ulcers healing and p80/p130Amot was the protective factor of DFU healing.ConcIusion:Expression level of Amot in 4 groups of patients with diabetic foot ulcers was lower than that of control group and p-80amot was dominant in 5 groups.Diabetes and PVD of DFU could reduce the expression level of Amot and PVD had a more important influence on Amot than diabetes.Control of blood glucose and reconstruction of vessel may contribute to the expression of amot and ulcer healing.The expression downregulating of Amot could speed down the DFU heling.p80/p130Amot was the protective factor of DFU healing,change of it in pathogical conditions may disturb the procession of angiogenesis and hinder DFU healing.
