Objective:This study was to investigate the anticancer and analgesic effect of UA nanoliposome and explore the possible mechanism and the relationship bettwen these two kinds of effects.Methods:The effect of UA nanoliposome on proliferation of HepG-2 cells was determined by MTT assay.The morphological changes were observed by Hoechst 33258 fluorochrome staining and electron microscope.Apoptosis rate were analysed by flow cytometry.The molecular mechanisms involved in the effects of UA nanoliposome on HepG-2 cells were studied by western blot.The effect on the survival time was testified in H22 ascite liver cancer mice.Models of writhing test and hot palate test and formalin paw edema experiment were used to testify the analgesic effect of ursolic acid.Results:MTT assay showed that UA nanoliposome had a anti-proliferation effect on HepG-2 cells in a dose-dependant manner.Apoptosis morphological changes about chromatic agglutination and nuclear condensation were detected by Hoechst 33258 fluorochrome staining and electron microscope in UA nanoliposome treated HepG-2 cells.Apoptosis rate of HepG-2 cells treated with Control and 30,90,150?mo/L UA nanoliposome for 24 hours were 9.03%?10.18%?14.78%?15.88%,(P<0.05)Respectively,The Bax proteins were upregulated and Bcl-2 proteinis was downregulated by western blot.Treatment of H22 ascites mice with UA nanoliposome significantly prolong the survival time of H22 ascites mice.Comparing with the negative control group,UA nanoliposome can extend the time of the pain threshold,reduce the number of writhing and inhibit the shaking action of mice.Conclusion:UA nanoliposome has a significant effect on inhibiting proliferation,inducing apoptosis of HepG-2 cells in vitro.Reducing the expression of Bcl-2 and increasing the expression of Bax may be its mechanism.It also can prolonge the survival time of H22 ascites mice.Meanwhile,UA nanoliposomes have weak analgesic activity.
