Determination Of The Effective Components' Contents In Tonic Chinese Patent Medicines By Multi- Wavelength HPLC

As we know,some compositions containing in prescription medicine of Chinese patent medicines have obvious pharmacological effect.Gallic acid in Guishao Dihuang pill,Mingmu Dihuang pills and Maiweidihuang pills has the effect of antibacterial,antiviral,antitumor.Paeoniflorin can dilate blood vessels,and has the sedative effects of analgesic,anti-inflammatory,anti ulcer and diuretic.Loganin has anti-cancer,antibacterial,anti-inflammatory,antitussive and expectorant effect.Ferulic acid can inhibit platelet aggregation,enhance the activity of prostaglandin,ease vasospasm and ease pain.Paeonol has the effects of antipyretic and analgesic,sedative,hypnotic,antibacterial,anti-inflammatory,antioxidant,lowering blood pressure and inhibit the allergic reaction.Ursolic acid has the functions of sedation,anti-inflammatory,antibacterial,anti-oxidation,anti ulcer,reducing blood sugar and so on.The contents of these active components have significantly effect to the efficacy of the drug.So it has important implication for how to determine the contents of these effective components easily,quickly and exactly.The multi-wavelength HPLC method was established to determinate the effective component contents of three kinds Tonic Chinese Medicine guishaodihuang pills,mingmudihuang pills and maiweidihuang pills.And the effective component contents of medicines from different manufacturers were compared.It provided basis for the quality standard of guishaodihuang pills,mingmudihuang pills and maiweidihuang pills.The research composed of three parts as follows.Part one Simultaneous determination of six components in Guishao Dihuang pills by HPLC methodObjective: To establish multi-wavelength HPLC analytical method for the simultaneously determination of Gallic Acid,5-hydroxymethylfurfural,Paeoniflorin,Loganin,Ferulic Acid and Paeonol in guishaodihuang pills.Methods: An Agilent Eclipse XDB-C18 column(5 ?m,150 mm×4.6 mm)was used.The mobile phase A was 0.1% phosphoric acid aqueous solution and the mobile phase B was methanol.Gradient elution mode was used.The flow rate were 1.0mL/min with the column temperature of 30?.The detection wavelength was 230 nm for Paeoniflorin and Loganin,275 nm for Gallic acid,5-hydroxymethylfurfural and Paeonol,and 320 nm for Ferulic Acid.Results: The peak areas and concentrations had good linear relationships for Gallic acid,5-hydroxymethylfurfural,Paeoniflorin,Loganin,Ferulic Acid and Paeonol in the ranges of 6.70-107.16 ?g/mL(r=0.999 9,n=5),5.84-93.39 ?g/mL(r=0.999 9,n=5),18.0-289.00 ?g/mL(r=0.999 9,n=5),5.73-91.66 ?g/mL(r=0.999 9,n=5),0.38-6.00 ?g/mL(r=0.999 8,n=5)and 13.18-210.90 ?g/mL(r=0.999 9,n=5),respectively.The average recoveries of the 6 kinds compositions were 100.14%,100.17%,100.23%,100.09%,100.19% and 100.08%,respectively.And RSD(n=9)of them were 0.72%,0.64%,0.51%,0.85%,0.93% and 0.65% respectively.Conclusion: The method is accurate and reliable,the reproducibility is good and the operation is simple and fast.So it can be used for the quality control of guishaodihuang pills.Part two Simultaneous determination of six effective components in mingmudi-huang pills by HPLC method.Objective: To establish multi-wavelength HPLC analytical method for the simultaneously determination of Gallic acid,5-hydroxymethylfurfural,Paeoniflorin,Loganin,Ferulic Acid and Paeonol in mingmudihuang pills.Methods: An Agilent Eclipse XDB-C18 column(5 ?m,150 mm×4.6 mm)was used.The mobile phase A was 0.1% phosphoric acid aqueous solution and the mobile phase B was methanol.The gradient elution mode 9,n=5),respectively.The average recoveries of the 6 kinds compositions were 100.11%,100.10%,100.02%,100.16%,100.19% and 100.14%,respectively.And RSD(n=9)of them were 0.68%,0.70%,0.95%,0.87%,1.16% and 0.41% respectively.Conclusion: The method is accurate and reliable,the reproducibility is good and the operation is simple and fast.So it can be used for the quality control of mingmudihuang pills.Part three Simultaneous determination of five effective components in maiweidihuang pills by HPLCObjective: To establish multi-wavelength HPLC analytical method for the simultaneously determination of Gallic acid,5-hydroxymethylfurfural,Paeoniflorin,Loganin,Ferulic Acid and Paeonol in maiweidihuang pills.Methods: An Agilent Eclipse XDB-C18 column(5 ?m,150 mm×4.6 mm)was used.The mobile phase A was 0.1% phosphoric acid aqueous solution and the mobile phase B was methanol.Gradient elution mode was used.The flow rate were 1.0mL/min with the column temperature of 30?.The Gallic acid,5-hydroxymethylfurfural,Paeoniflorin,Loganin and Paeonol in the ranges of 6.70-107.16 ?g/mL(r=0.999 9,n=5),5.84-186.77 ?g/mL(r=0.999 9,n=6),9.04-144.60 ?g/mL(r=0.999 9,n=5),5.73-91.66 ?g/mL(r=0.999 9,n=5)and 13.19-211.00 ?g/mL(r=0.999 9,n=5),respectively.The average recoveries of the 6 kinds compositions were 100.15%,100.33%,100.10%,100.40% and 100.34%,respectively.And RSD(n=9)of them were 0.92%,0.67%,0.77%,0.72% and 0.35%,respectively.Conclusion: The method is accurate and reliable,the reproducibility is good and the operation is simple and fast.So it can be used for the quality control of maiweidihuang pills.
