To establish a multi-wavelength HPLC method for the simultaneous determination of Huanglianshangqing tablets, Zhizijinhua pills, Qingwei huanglian pills and Lianqiaobaidu pills, four kinds of Chinese medicine compound preparation of effective component content, and the comparison among different manufacturers of drug quality,the research composed of four parts as follows. Part 1 Determination of content of effective components of Huangli-anshang qing tablets from different manufacturers by HPLCObjective: To establish a HPLC method for the determination of geniposide, berberine hydrochloride and baicalin in Huanglianshang qing tablets simultaneously, and the determination of emodin. And to determine the contents of four components in Huanglianshangqing tablets produced by four manufacturers.Methods: An Agilent Eclipse XDB-C18 column(5 μm, 150 mm×4.6 mm)was used.The mobile phase were composed of 0.1% phosphoric acid aqueous solution(0.1% triethylamine)- methanol to determine the three(geniposide, berberine hydrochloride and baicalin)components simultaneously by gradient elution mode; the mobile phase were composed of 0.1% phosphoric acid aqueous solution and methanol(20:80) to determine the emodin. The flow rate were 1.0 m L/min with the column temperature 30 ℃,The detection wavelength were chosen as 238 nm for、230 nm for、278 nm for and 254 nm for.Results: The concentration ranges of Geniposide, berberine hydrochloride, baicalin and emodin were in 1.95-125.00 μg/m L(r =0.999 9, n=5),1.25-80.00 μg/m L(r =0.999 9, n=5), 3.125-200.00 μg/m L(r=0.999 9,5 n=5), 6.25-80.00 μg/m L(r =0.999 8,n=5) respectively with a good linear relationship between peak area. The average recoveries were 99.88%, 100.2%, 100.8% and 100.3%, RSD(n=9)were 0.92%, 0.92%, 1.18% and 0.61% respectively.Conclusion: The method is accurate and reliable, the reproducibility is good and the operation is simple and fast, which can be used for the quality control. Part 2 Simultaneous determination of four effective components in Zhizij-inhua pills by HPLCObjective: To establish a HPLC method for the determination of chlorogenic acid, geniposide, berberine hydrochloride and baicalin in Huanglianshangqing pills simultaneously. And to determine the contents of four components in Zhizijinhua pills produced by three manufacturers.Methods: An Agilent Eclipse XDB-C18 column(5 μm,150 mm×4.6 mm)was used.The mobile phase were composed of 0.5% phosphoric acid aqueous solution and methanol by gradient elution mode at a flow rate of 1.0 m L/min with the column temperature 30 ℃.The detection wavelength were chosen as 324 nm for, 238 nm form 345 nm for and 278 nm for.Results: Chlorogenic acid, geniposide, berberine hydrochloride and baicalin concentration respectively in 1.75-28.00 μg/m L(r=0.999 9,n=5), 3.00-48.00 μg/m L(r=0.999 9,n=5), 1.188-19.00 μg/m L(r =0.999 8,n=5), 22.50-360.00 μg/m L(r=0.999 9,n=5) and a good linear relationship between peak area. The average recoveries were 99.93%, 100.3%, 100.7% and 100.6%, RSD(n=9) were 0.64%,0.63%,1.25% and 0.63% respectively.Conclusion: The method is accurate and reliable, the reproducibility is good and the operation is simple and fast,which Can be used for the quality control. Part 3 Simultaneous determination of five effective components inQingwei huanglian pills by HPLCObjective: To establish a HPLC method for the determination of geniposide, paeoniflorin, berberine hydrochloride, paeonol and baicalin in Qingweihuang lian pills simultaneously,And to determine the contents of five components in Qingweihuanglian pills produced by three manufacturers.Methods: An Agilent Eclipse XDB-C18column(5 μm,150 mm×4.6 mm)was used.The mobile phase were composed of 0.1% phosphoric acid aqueous solution and methanol by gradient elution mode at a flow rate of 1.0 m L/min with the column temperature 30 ℃. The detection wavelength were chosen as 238 nm for(geniposide), 230 nm for(paeoniflorin and berberine hydrochloride), 276 nm for(paeonol and baicalin).Results: Geniposide, paeoniflorin, berberine hydrochloride,paeonol and baicalin concentration respectively in 4.375-70.00 μg/m L(r=0.999 9, n=5), 2.85-45.60 μg/m L(r=0.999 9, n=5), 10.00-160.00 μg/m L(r=0.999 8, n=5), 0.75-12.00 μg/m L(r=0.999 8, n=5), 10.00-160.00 μg/m L(r=0.999 9, n=5)and a good linear relationship between peak area. The average recoveries were 99.38%, 99.77%, 99.23%, 98.81% and 99.96%, RSD(n=9) were 0.86%, 1.03%, 1.30%, 1.37% and 0.06 % respectively.Conclusion: The method is accurate and reliable,the reproducibili-ty is good and the operation is simple and fast, which Can be used for the quality control. Qingweihuanglian pills from different manufacturers, as the preparation process and raw material quality are different, there are large differences in product quality. Part 4 Simultaneous determination of five effective components in Lian-qiaobaidu pills by HPLCObjective:To establish a HPLC method for the determination of chlorogenic acid, paeoniflorin, ferulic acid, berberine hydrochloride and baicalin in Lianqiaobaidu pills simultaneously. And to determine the contents of five components in Lianqiaobaidu pills produced by two manufacturers.Methods: An Agilent Eclipse XDB-C18 column(5 μm,150 mm×4.6 mm)was used.The mobile phase were composed of 0.5% phosphoric acid aqueous solution(0.03% triethylamine) and methanol by gradient elution mode at a flow rate of 1.0 m L/min with the column temperature 30 ℃,The detection wavelength were chosen as 230 nm for(paeoniflorin and berberine hydrochloride), 324 nm for(chlorogenic acid and ferulic acid) and 278 nm for(baicalin).Results: chlorogenic acid,paeoniflorin,ferulic acid,berberine hydrochloride and baicalin concentration respectively in 9.10-154.60 μg/m L r=0.999 9, n=5),1.90-30.40μg/m L(r=0.9997,n=5), 0.51-8.16 μg/m L,(r=0.9997, n=5), 6.50-104.00 μg/m L(r =0.999 8, n=5), 9.75-156.00 μg/m L( r=0.9999, n=5) and a good linear relationship between peak area. The average recoveries were 100.5%, 100.5%, 100.0%, 100.7% and 100.0% RSD( n=9) were 0.93%, 0.90%, 0.86%, 0.82% and 0.62% respectively.Conclusion: The method is accurate and reliable,the reproducibility is good and the operation is simple and fast,which Can be used for the quality control. Lianqiaobaidu pills from different manufacturers, as the preparation process and raw material quality are different, there are large differences in product quality.
