Objective: To observe the influence of autologous blood storage on peripheral blood routine and serum IL-3,IL-11 in the New Zealand rabbits.To explore the effect of preoperative autologous blood donation on the bone marrow hematopoietic cells in the acute hemorrhagic shock rabbits.Comprehensive evaluate the safety and efficacy of preoperative autologous blood donation.Methods: To elective clean and healthy twenty-four male New Zealand rabbits,weighing1.9-2.4kg,were randomly divided into 4 groups?n=6 each?,using a random number table:control group?group CON?,allogeneic blood transfusion group?group ABT?,autologous blood storage group?group ABS?and predeposit autotransfusion group?group PAT?.ABS and PAT groups were collected and stored autologous blood for 3 times before simulate shock,every time collect 12% of the total blood volume of blood,once a week and 72 hours preceding shock.Femoral artery of group CON and group ABS were separated,but don't simulate shock,after establish the rabbit model of acute hemorrhagic shock,group ABT transfused the blood stored by group ABS,group PAT transfused the autologous blood collected before shock,input Sodium Lactate Ringer's to achieve recovery standard if necessary.Dynamic monitoring mean arterial pressure?MAP?during the Simulation of hemorrhagic shock and resuscitation.The peripheral blood routine and serum IL-3,IL-11 of four groups of rabbits were tested before blood storage?T1??after blood storage?T2??before hemorrhagic shock?T3?,resuscitation 0 min?T4?,resuscitation 24h?T5?.The bone marrow nucleated cell of rabbits was counted at resuscitation 24 h,the marrow hematopoietic cell cycle was examined 24 h after resuscitation by flow cytometry,and smear the bone marrow,microscopy after staining.Results:?1?Four groups of rabbits basis MAP was no significant difference?P>0.05?.Compared with the base level MAP,MAP of group ABT and group PAT were significantly decreased after shock?P<0.05?,then returned to the pre-shock levels at resuscitation 0.?2?Compared with T1,Hb concentration of group ABT were significantly reduced at T4?P<0.05?;then returned to the pre-shock levels at T5.Compared with group CON,Hb concentration of group ABS were significantly decreased,and remained at a low level at resuscitation 24h?P<0.05?.Compared with group CON,Hb concentration of group PAT were significantly decreased at T2 and T3?P<0.05?,but was no significant difference at T4,T5.Compared with group ABT,RET% of group PAT and group ABS were significantly increased after blood storage?P<0.05?,and still at a high level after 24 h of recovery?P<0.05?.There were no significant difference between four groups of rabbits WBC,PLT at different time?P > 0.05?,there were no significant difference between groups at the same time point?P>0.05?.Compared with group CON,RBC of group ABS and group PAT reduced at T2,T3?P<0.05?,group ABS still at a low level at T4,T5.Compared with T1,RBC of group PAT decreased at T2,T3?P<0.05?,returned to the baseline level at T4?P>0.05?;RBC of group ABS reduced at T4,still below the basic level at T5?P<0.05?.?3?Compared with group CON,group ABT,serum IL-3,IL-11 of group ABS and group PAT was significantly increased after storage of blood?P<0.05?.Compared with T1,serum IL-3,IL-11 of group ABS and group PAT increased after storage of blood,and still at a high level after 24 h of recovery?P<0.05?.?4?Compared with group CON and group ABT,the number of bone marrow nucleated cell was significantly increased in group ABS and PAT?P<0.05?.?5?Compared with group CON and group ABT,the marrow hematopoietic cell cycle of group ABS and group PAT G0/G1 phase was significantly decreased,the S phase,the G2/M phase cell proportion obviously advanced?P<0.05?.Compared with group CON,the marrow hematopoietic cell cycle of group ABT G0/G1 phase was significantly increased,the S phase,the G2/M phase cell proportion obviously reduced?P < 0.05?.?6?Bone marrow smear observe that ratio of myeloid granulocyte/ nucleated red blood cell of group ABS and group PAT decreased,nucleated immature red blood cells increased obviously,bone marrow cells proliferated actively and can visible bone marrow hematopoietic island and platelet aggregation.Conclusion:?1?Autologous blood storage can reduce the content of peripheral blood Hb,RBC,but can quickly recover those to the basic level after the transfusion,and has more effective than allogeneic blood transfusion.?2?Preoperative autologous blood donation can stimulate the bone marrow hematopoietic cell from the stationary phase to proliferation,increase the bone marrow nucleated cell,enhance the percentage of reticulocyte and content of IL-3,IL-11,accelerate the early recovery of peripheral blood.?3?Allogeneic blood transfusion inhibit the bone marrow hematopoietic cell from the stationary phase to proliferation,effects the early recovery of Hb and RBC.In short,preoperative autologous blood donation is a safe,effective blood conservation methods.
