Objective To evaluate the effect of stored autologous blood transfusion on bone marrow hematopoietic function and hematopoietic stem cells by observing the changes in the number and activity of bone marrow CD34+cells after storage autologous blood transfusion.Methods 30 New Zealand rabbits were randomly divided into 5 groups with 6 rats in each group.Group A,blank control group;Group B,surgery alone group;Group C,simple blood collection group;Group D,storage type autologous whole blood group;Group E,storage autologous component blood transfusion group.Bone marrow blood samples were taken before operation(T2),6 hours after operation(T3),and 24 hours after operation(T4).The percentage of CD34+cells in each group was counted by flow cytometry,and the cell cycle was detected.Relative expression of telomere DNA in CD34+cells and peripheral blood samples were taken before autologous blood storage(T1)and 24 hours after surgery(T4),and peripheral blood reticulocyte content was detected by using a blood cell analyzer.Results There was no significant difference in body weight,total operation time and total anesthetic dose between the rabbits(P>0.05).At the time of T3 and T4,the percentage of CD34+cells in each experimental group was significantly higher than that in the blank control group(group A).The number of cells in G1 phase was significantly decreased,and the expression of telomeric DNA was significantly increased(P<0.05).At T3,T4 The percentage of CD34+cells in group D and group E was significantly higher than that in group C.The cells in G1 phase were significantly lower than those in group C(P<0.05).The expression of telomere DNA was significantly higher than that in group C.At group T3 and T4 The percentage of CD34+cells was significantly lower than that of group D.The cells in G1 phase were significantly higher than those in group D(P<0.05),and the expression of telomeric DNA was significantly lower than that in group D.At the time of T4,the percentage of reticulocytes in group B and group C was significantly higher than that in group E and group D(P<0.05).The percentage of reticulocytes in group E was significantly higher than that in group D(P<0.05).Conclusion Preoperative autologous blood collection can enhance the hematopoietic function of bone marrow.Compared with stored autologous whole blood,the stored autologous component blood transfusion has less influence on the hematopoietic function of bone marrow,and the number of recovered peripheral blood cells is larger.The storage type autologous component is excellent in blood transfusion.In storage of autologous whole blood.
