| Objective: Basd on researching the source,character,microscopy,roution inspection,extract,content determination and fingerprint,we establish the quality standard of Zhuang medicine of Lindernia ruellioides(Colsm.)Pennell.Methods: Through the origin identification,character identification,microscopic identification and physical and chemical identification methods to systematically qualitative study on pharmacognosy of Lindernia ruellioides(Colsm.)Pennell.According the methods stated in the fourth volume of Chinese Pharmacopeia(2015 edition)to research its examination of limited,including moisture,total ash,acid-insoluble ash and extract and so on.According to the method of Chinese Medicine Chemistry experiment to forecast the chemical composition in Lindernia ruellioides(Colsm.)Pennell.With acteoside as the reference substance,determining the content in Lindernia ruellioides(Colsm.)Pennell of different habitats by HPLC and confirming its limit of acteoside.At the same time,with acteoside as the reference peak,researching the fringerprint of Lindernia ruellioides(Colsm.)Pennell by the soft of traditional medicine chromatographic fingerprint similarity evaluation system 2012 edition.Results: 1.The pharmacognosical characteristics of origin identification,macroscopic identification,microscopic identification,physical and chemicalidentification are remarkable and can be used as the basis of identifying Lindernia ruellioides(Colsm.)Pennell.2.The moisture in the Lindernia ruellioides(Colsm.)Pennell between 8.24 % and 11.76 %,all bellowed 12.0 %,so it couldn’t be more than 13.0 %;the total ash between 11.62 % and 17.36 %,and most of bellow 16.0%,so it couldn’t be higher than 18.00%;the acid-insoluble ash between 1.55 % and 8.28 %,but most of bellow 7.0 %,so it should not exceed 9.0 %;the content of ethanol extracts between 17.27 % and27.02 %,all above 17.0 %,so it shall not be less than 15.0 %.3.It shows that phenolic compounds,triterpenoid compounds,steroids,lactones,polysaccharides,amino acids,volatile oil components are included in Lindernia ruellioides(Colsm.)Pennell.4.The optimal extraction technology for Lindernia ruellioides(Colsm.)Pennell was that adding 70 % methanol 20 ml in the sample with 0.4 g,ultrasonic processing two times with each time 30 minutes.Between 0.0192~3.84 μg,the content of acteoside has good linear relationship with the peak area(r = 0.99999).The average sample recovery rate was 103.59 % RSD =2.88 %;The acteoside contents of each batch Lindernia ruellioides(Colsm.)Pennell between 0.23 % and 0.94 %.Based on the most contents are more than 0.20 %,the content of acteoside shall not be less than0.20 %(2.00 mg·g-1).5.The HPLC fingerprint and chromatographic conditions of Lindernia ruellioides(Colsm.)Pennell were established preliminarily.Selecting 15 characteristic peaks with acteoside as reference peak,the methodology is up to standard.And the similarity of the different batches of samples was above 0.96.Conclusion: Based on above evaluations,medicinal material quality standard of Lindernia ruellioides(Colsm.)Pennell was initially estabished,it can lay a foundation for controling safe,effective and quality of Lindernia ruellioides(Colsm.)Pennell on the clinical application. |
