Objective: Ethyl acetate extract and n-butanol fraction,extracted from the whole herbs of Ixora chinensis Lam.,are used to investigate the chemical compositions and pharmacological effects in this study.This work can lay a foundation for the future activity compounds screening(to reveal the material foundation of its bio-activities)and pharmacological mechanism study in future,and further to provide an evidence for exploitation and utilization of this medicinal plant.Methods: The crude powder of the whole plant was extracted with ethanol and water by percolation to give a total ethanol extract and water extract.The ethanol extract was suspended in water and extracted by petroleum ether,ethyl acetate and n-butanol subsequently to obtain petroleum ether fraction,ethyl acetate fraction,n-butanol and the remaining parts of water.The methods of silicagel column chromatography,polyamide column chromatography,macroporous resin chromatography,Sephadex LH-20 column chromatography and recrystallization,were used to isolate and purify chemical constituents from the ethyl acetate fraction and n-butanol fraction.The spectroscopic meth-ods(EI-MS,ESI-MS,1H-NMR,13C-NMR,DEPT)and physical and chemical tests were used to identify the structures of the obtained compounds.To the known com-pounds,the literature data were consulted.Meanwhile,ethyl acetatefractio-n,n-butanol fraction water fraction and Ixora Chinensis Lam.ester acid were used to detect their antitumor activity in human lung cancer cell line A549,human hepatoma cell line SMMC-7721 by cytotoxicity assay(MTT method).In addition,through one-step detection of mouse liver tissue malo-ndialdehyde(MDA)content method,ethyl acetate-fraction,n-butanol fra-ction and water fraction were used to observe their lipid peroxide(LPO)inhibitory activity on rat liver.Data obtained were processed by SPSS 17.0software.Results: Eleven compounds were isolated from the ethyl acetate and n-butan-ol fraction.Andten of them have been identified as monohydroxysterols(22E,24S)-24-methylcholesta-5,22-dien-3?-ol(1),scopoletin(2),palmitic acid(3),ursolic acid(4),salicylic acid(5),?-daucosterol(7),kaempferol-3-O-glucos(6?1)-O-rhamnose-7-O-rhamnoside(8),1-butyl-6-methyl glucoside(9),kaemmpfe-rol(10),caprolactam(11),mannitol(12).In antitumoractivity,Ixora Chinesis ex-tracts have a certain extent on liver cancer cells and lung cancer cells,and the ethyl acetate extract and Ixora chinensis Lam.ester acid was more pronounced.The half inhibitory concentration(IC50)of the ethyl acetate extract of A549 and SMMC-7721 were 0.395g·l-1 and 0.891g·l-1.The half inhibitory concentration(IC50)of Ixora Chinensis Lam.ester acid of A549 were 0.497g·l-1.In the LPO inhibitory activity aspect,ethyl acetate,n-butanol and water parts have a certain effect of anti-lipid,and the low-dose group of ethyl acetate and n-butanol ext-racts were better on anti-lipid peroxidation effect.Conclusion:The above compounds were mainly triterpenoids,steroids and flavonoids.Compounds1-5,7-11 were isolated from this plant for the first time.However,compound12(mannitol)has been reported.Ixora Chinensis La-m.has some anti-tumor and anti-lipid peroxidation inhibition.
