Objective To observe the effect of ascorbic acid preconditioning on the glomerular endothelial polysaccharide protein complex in diabetic rats after renal ischemia and reperfusion.Methods 32 healthy SD rats were randomly divided into 4 groups, group A with normal blood glucose, groupB?C and D are in diabetic rats, group A and group B as control group; group C and group D were exposed to ischemia- reperfusion treatment;group D were given ascorbic acid 200 mg/kg- 1/d- 1 i. P. for 15 days. To observe the changes of blood glucose, blood urea nitrogen(BUN), creatinine(SCR),malondialdehyde(MDA), superoxide dismutase enzyme(SOD)at 30 min before operation(T1), in front of ischemia(T2), ischemia for 30min(T3), reperfusion6h(T4)and reperfusion 24h(T5),test the expression of caspase- 3(caspase-3),cytochrome c(cytc), polysaccharide protein complexes after reperfusion for 24 h.Results(1)In group C and D, the levels of BUN ?SCr and MDA at T3, T4 and T5 were significantly higher than those in group A and B, with T4 as peak, SOD level was significantly lower than group A and B, and T4 was low(P<0.05).(2)The levels of BUN? SCr?MDA at T3, T4, T5 in group D were significantly decreased,SOD levels were significantly increased, caspase-3 and cyt C expression were significantly reduced and a significant increase in the thickness of the glomerular endothelial polysaccharide protein complex, renal tissue injury significantly improved(P < 0.05) when compared with group C.Conclusion Ascorbic acid preconditioning can reduce the injury of renal ischemia reperfusion injury in diabetic rats by reducing the injury of glomerular endothelial protein complex after renal ischemia reperfusion in diabetic rats.
